Ovarian cancer is one of the three major malignant tumors of the female reproductive system,and its specific pathogenesis is still unclear,and the therapeutic effect is not satisfactory.One of the main reasons for the unsatisfactory treatment of ovarian cancer is the occult symptoms of ovarian cancer,so most patients are in advanced stage at the time of initial diagnosis.There is currently no effective screening and evaluation method for ovarian cancer.On the other hand,the treatment of ovarian cancer is also limited,and the development of new therapeutic drugs is imminent Metabolomics is the qualitative and quantitative analysis of relatively low molecular weight metabolites of a certain organism or cell,reflecting the subject's response to environmental conditions or genetic changes,revealing the overall metabolic changes of the organism.Metabolomics is closely related to drug efficacy,safety assessment,mechanism of action,and rational drug use.In this paper,we used animal model combined with liquid chromatography-mass spectrometry and chemometric pattern recognition method to conduct a comprehensive metabolomics analysis of ovarian cancer-bearing nude mice serum to identify differential metabolites for ovarian cancer To investigate the therapeutic effect of oleuropcin on ovarian cancerThe research content of this thesis is divided into the following two parts:1.The ultra-high performance liquid chromatography-mass spectrometry technique used in this study and the related multivariate statistical analysis methods required for this study are introduced.The materials,specific steps and operation methods of this experiment are described in detail2.To determine the inhibitory effect of oleuropein on ovarian cancer by measuring the body weight of ovarian cancer-bearing nude mice and the volume of subcutaneous xenografts.To establish the serum metabolic profile of ovarian cancer-bearing nude mice,and to establish a correlation model based on multivariate statistical analysis methods,and to screen out differential metabolites related to this study,including pseudouridine,phenylacetylglycine,arginine,cinnamaldehyde,6-aminocaproic acid,docosahexaenoic acid.It is concluded that oleuropein can improve intestinal flora and express anti-tumor activity by affecting phenylalanine metabolism,and inhibit tumor by affecting arginine metabolic pathway;At the same time,it was found that DHA,cinnamaldehyde and 6-aminocaproic acid may be important components in the anti-tumor effect of oleuropein;pseudouridine may be a potential biomarker for the diagnosis and prognosis of ovarian cancer.
