Objective:This experiment is to observe the expression of protein ip-10,KLK5 and MIG in the serum and tissues of laryngeal carcinoma,and to explore its correlation with laryngeal carcinoma,providing a scientific basis for early diagnosis of laryngeal carcinoma.Methods: The study is divided into two parts.Part I:To collect 80 cases of blood samples of patients in the First Hospital Affiliate d to south of China University?Hengyang Centre Hospital?Nanhua Hospital Affiliated to south of China University in July 2016-Octob er 2017,There were 21 cases in the untreated group,29 in the tre atment group,20 in the laryngeal polyp group and 10 in the contro l group?Using ELISA method respectively in four groups to determi nation of protein concentration levels in serum IP-10,KLK5 and M IG.Comparison of three kinds of inflammatory factors in each group,finally using statistical methods to analyze the statistical results.Part II:to collect 50 cases of laryngeal malignancy and 20 cases of paracarcinoma tissue between May 2013 and December 2015 in the Sec ond affiliated Hospital of south of China University.Expression of MIG and KLK5 in samples of laryngeal cancer tumor and para-carc inoma tissue by immunohistochemistry(SP),and follow up.Analysis of its expression in laryngeal cancer cells,to compared para-carcino ma tissue cells in the larynx.and analyzing the relationship betwee n the expression and gender,age,smoking history,TNM staging,di fferentiation degree,lymph node metastasis,the prognosis and other related factors in laryngeal malignant tumor patients.Results:1.The serum expression level of ip-10 protein in the control group,polyp group,untreated group,and treatment group was 561.65 ± 371.65pg/ml,751.89 ± 426.06pg/ml,767.85 ± 409.97pg/ml,704.10 ± 548.99pg/ml.The positive rate was 10.00%,9.52% and 13.79% in the polyp group,untreated group and treatment group.The expression in each group was not statistically significant(P>0.05),In untreated group,the concentrations of phase I?II and III?IV were 779.35±355.11pg/ml?757.40±471.59pg/ml;The positive rate was 10.00% and 18.18%.there was no statistical significance in different clinical stages(P>0.05).2.The serum expression level of KLK5 protein in the control group,polyp group,untreated group and treatment group was 371.78 ± 226.69pg/ml,518.40 ± 221.71pg/ml,738.78 ± 356.77pg/ml,542.51 ± 248.26pg/ml.The positive rate was 5.00%,33.33% and 13.79% in the polyp group,untreated group and treatment group,and the expression in each group was statistically significant(P < 0.05).In untreated group,the concentrations of stage I?II and III?IV were 629.04±290.93pg/ml?838.55±394.22pg/ml;The positive rate was 20.00% and 36.36%,there was no statistical significance in different clinical stages(P>0.05)?KLK5 had a positive rate of 10.00% and 6.00% in the adjacent tissues and throat cancer tissues.KLK5 expression was not statistically significant in laryngeal cancer tissue(P> 0.05).3.The serum expression levels of MIG protein in the control group,polyp group,untreated group and treatment group were 115.36 ± 48.60pg/ml,143.61 ± 84.53pg/ml,251.92 ± 140.22pg/ml,157.01 ± 69.62pg/ml.The positive rate was 15.00%,61.90% and 31.81% in the polyp group,untreated group and treatment group,and the expression in each group was statistically significant(P < 0.05).In untreated group,tthe concentrations of stage I?II and III?IV were 181.67±107.68pg/ml?315.78±139.51pg/ml;The positive rate was 30.00% and 72.73% respectively.The expression was statistically significant in different clinical stages(P < 0.05).MIG had a positive rate of 5.00% and 62.00% in the adjacent tissues of cancer,and the expression of MIG in laryngeal cancer was statistically significant(P < 0.05).The expression of MIG in laryngeal carcinoma was not related to age,gender,smoking history,T staging(P > 0.05),The degree of differentiation and lymph node metastasis were statistically significant(P < 0.05).The survival rate of MIG expression positive patients was not statistically significant(P > 0.05).Conclusions:1.The expression of KLK5 protein in the serum of laryngeal carcinoma was increased.2.The expression of MIG protein in the serum of laryngeal cancer and the increase of positive rate were related to the clinical stage of laryngeal cancer.The expression of MIG protein in laryngeal carcinoma is related to the degree of differentiation and lymph node metastasis.
