Objective: The purpose of this study is to observe the changes of thyroxine receptor ?1(TR?1)and SUMO protein expression during hepatocyte lipid transformation,to investigate whether sumoylation of TR ?1 participates in the process of hepatic lipid deposition,so as to provide a new target for clinical NAFLD pathogenesis.Methods:1.Immunohistochemistry confirmed the expression of TR?1 and SUMO in human liver tissue of NAFLD;2.Culture human normal liver cell WRL68 cells in vitro,induce WLR68 cell steatosis with different concentration of free fatty acid(FFA)(OA: PA=2:1),select the optimal induction concentration and time,and establish a fatty liver cell model;3.The WRL68 cells were divided into two groups: normal control group and FFA-induced fatty liver cell group.The levels of triglyceride and cholesterol were measured by biochemical kit at 6H and 24 H,and the lipids of liver cells were evaluated by oil red staining.Degree ofdeposition;4.The expression of TR?1 and SUMO in hepatocytes was detected by Western blot.Results:1.The expression of TR TR?1,SUMO1 and SUMO3 in liver tissue of NAFLD patients was significantly higher than that of normal subjects.2.The results of lipid deposition in hepatocytes showed that WRL68 cells were treated with different concentrations of FFA(0.0mM ?0.25mM?0.5mM)for 6H?24H and 48 H,to induced hepatocyte steatosis.The fatty liver cell model was successfully established.Combined with oil red staining results and intracellular TG and TC values.The best induced concentration of FFA was 0.5 mM,and the best time for phenotypic observation was observed 24 H after induction.3.Western blot analysis of WRL68 cells showed that TR?1 and SUMO1?SUMO3 protein expression in FFA-induced group were higher than those in control group,and the expression level increased with the increase of induction time.Conclusion:1.The expression of TR?1 and SUMO1?SUMO3 protein in NAFLD hepatic tissue increased.2.The expression of TR?1 and SUMO1 ? SUMO3 protein in the process of hepatocyte lipogenesis was increased,SUMO-modified TR?1involved in the process of hepatocyte lipogenesis.
