Dendrobium nobile Lindl.,a traditional Chinese medicine,has many functions such as enhancing immunity,protecting the gastrointestinal tract,decreasing blood glucose,decreasing blood lipid and so on.In this study,the extract of Dendrobium nobile Lindl.which was used as the main ingredient,and the extract of Lycium barbarum and Radix Puerariae lobatae was selected to form a compound.The ratio of the compound was optimized based on the principle of uniform design and the compatible effect was studied.Then a stable compound oral liquid was developed and its immunoenhancement and stomach protection efficacy were evaluated.The research content is as follows:1.Ingredient of the oral liquid was selected and the ratio of the compound was optimized.The phagocytosis ability of RAW264.7 macrophages and the proliferation rate of human gastric epithelial cells(GES-1)damaged by ethanol were used to select the ingredient,and the extract of Dendrobium nobile Lindl.,the extract of Lycium barbarum and the extract of Radix Puerariae lobatae was selected.Based on the principle of uniform design,the ratio of the oral liquid compound was obtained,Dendrobium nobile Lindl.extract:Lyciurm barbarum extract:Radix Puerariae lobatae extract was 5:3:3.2.The immune-enhancing and gastric protective effects of extracts of Dendrobium nobile Lindl,Radix Puerariae lobatae,Lycium barbarum and compound extracts were determined in vitro.To explore the compatible effect of the compound,the proliferation rate of splenic lymphocytes,RAW264.7 cell phagocytosis index,nitric oxide(NO)level and GES-1 cell proliferation rate were measured.The results showed that the compound could significantly promote the proliferation of lymphocytes at a concentration of 1000 ?g/mL,and the effect was better than that of Dendrobium nobile Lindl.extract and Lycium barbarum extract.At a concentration of 200 ?g/mL,the compound could significantly promot the phagocytic ability of RAW264.7 macrophages,and the phagocytic index was 1.96+0.01,which was higher than the phagocytic index of single extract;At 400 ?g/mL,the NO secretion ability of RAW264.7 cells treated with single and compound extracts was significantly enhanced,and the compound group had the highest NO level;At 400 ?g/mL,compared with the model group,single and compound extract treatment can relieved the damage of GES-1 cell viability induced by ethanol.The proliferation rate of GES-1 cell in the compound extract treatment group was 86.75%±4.56%,higher than single extract treatment group.The compound had the functions of enhancing immunity and stomach protection,and its effect was superior to the single extract,showing synergistic effect.3.The excipients optimization,physicochemical index determination and accelerated stability of oral liquid were studied.The excipient formula was xanthan gum 3.0 mg/mL,mogroside 2.0 mg/mL.The pH of the oral liquid was 4.0,the content of soluble solids was 6.6%,the content of polysaccharides was 13.97 mg/mL and puerarin was 3.88 mg/mL.The accelerated stability of oral liquid was tested for 3 months.During the experiment,the sensory quality,physical and chemical indexes and microbial indexes of the oral liquid met the product quality requirements.The stability of polysaccharides and puerarin,the main effective ingredients,was good and met the shelf life requirements of 24 months.4.The immunoregulatory activity of oral liquid was evaluated by BALB/c mice.Compared with the blank group,the spleen index and thymus index of mice in the high-dose oral liquid group increased significantly,respectively 4.46±0.63,3.61±0.45;the proliferation ability of spleen lymphocytes of mice in the low,medium and high dose oral liquid group increased significantly,the phagocytic index of mice in the high dose oral liquid group increased significantly to 5.28±0.43;The hemolytic plaques of the low,medium and high dose oral liquid pretreatment groups were significantly higher than those of the blank group.The oral liquid can enhance the specific and non-specific immunity of mice,which has good immunoregulatory effect.5.The stomach protection efficacy of oral liquid was evaluated by ethanol-induced gastric mucosal injury model in rats.With the increase of oral liquid dosage,the gastric mucosal damage induced by ethanol was gradually lightened,the area of hyperemia and ulcer were smaller,the exfoliation of epithelial mucosa was reduced,the extent of mucosal bleeding and the number of inflammatory cells were decreased.The gastric mucosal injury score in the high dose group was 15.5+4.18,which was significantly lower than that in the ethanol-induced model group.At the same time,medium and high dose oral liquid could effectively maintain the activity level of SOD,reduce MDA content,inhibit the release of gastric mucosal attack factor,and reduce the levels of inflammatory factors TNF-? and IL-1?,and improve epidermal growth factor(EGF)and prostaglandin E2 content.This study has confirmed that the oral liquid has an auxiliary protective effect on ethanol-induced gastric mucosal damage.
