OBJECTIVE:Bladder cancer is a common clinical disease of urinary system.It is the three most common tumors in urology with renal cancer and prostate cancer.In recent years,the incidence of bladder cancer has gradually increased,which has become a major potential threat to human health.This study was designed to investigate the effect of dibutyl phthalate(DBP)on the expression of Shh signaling pathway in bladder epithelial cells.To determine whether DBP promotes the development of bladder cancer by reducing the gene expression of Shh signaling pathway in bladder epithelial cells.To reveal the relationship between DBP and the occurrence of bladder cancer,and to clarify the molecular mechanism of DBP promoting the occurrence of bladder cancer.It is helpful to further improve people's awareness of DBP toxicity and prevention awareness,which is of great significance for strengthening people's health awareness,reducing the incidence of bladder cancer and improving people's health level.METHODS:1.Real-time PCR was used to detect the changes of Shh signaling pathway(Shh,Gli1,Gli2,Bmp4)gene expression in bladder epithelial cells and bladder cancer cells.To clarify the effect of DBP on Shh signaling pathway gene expression in bladder epithelial cells and bladder cancer cells.2.The expression of Shh signaling pathways(Shh,Gli1,Gli2,Bmp4)in bladder epithelial cells and bladder cancer cells was detected by Western Blot.To clarify the effect of DBP on the expression of Shh signaling pathway protein in bladder epithelial cells and bladder cancer cells.RESULTS:1.Real-time PCR results confirmed that DBP could significantly down-regulate the expression of Shh signaling pathways(including Shh,Gli1,Gli2,Bmp4)in bladder epithelial cells,with significant difference(P < 0.05).Compared with the normal bladder epithelial cells treated with DBP,the expression of Shh,Gli1,Gli2 and Bmp4 in the bladder cancer cells treated with DBP decreased more significantly(P < 0.05).2.Western blot results confirmed that DBP could significantly down-regulate the expression of Shh signaling pathways(including Shh,Gli1,Gli2,Bmp4)in bladder epithelial cells,with significant difference(P < 0.05).Compared with the normal bladder epithelial cells treated with DBP,the expression of Shh,Gli1,Gli2 and Bmp4 in the bladder cancer cells treated with DBP decreased more significantly(P < 0.05).CONCLUSIONS: DBP can reduce the expression of Shh,Gli1,Gli2 and Bmp4 in normal bladder epithelial cells and bladder cancer cells.The expression of Shh signaling pathway in bladder cancer cells treated with DBP was down-regulated more significantly than that in normal bladder epithelial cells.
