Purpose 1.To screen and identify a group of microRNAs with differential expression in serum of children with ADHD.2.Based on a group of microRNAs with different expression in children's serum of ADHD,to establish a logistic regression analysis model and to investigate the clinical reference value of this model for the diagnosis of ADHD.3.To investigate whether there is a change in serum differential microRNA expression in children with ADHD after treatment.Methods 1.From January 2015 to December 2016,160 children were collected as screening group,including 40 children with ADHD,120 healthy children.The microRNA profiles of children with ADHD and healthy children were obtained by using Agilent Human MiRNA Microarray V21.0 containing 2549 human microRNA probes.Statistically different microRNAs were screened and verified by qRT-PCR.2.From January 2017 to June 2017,160 children were collected as test group, including 80 children with ADHD,80 healthy children,30 children with pneumonia and 30 patients with vitamin D deficiency.The above selected microRNAs were subjected to independent RT-qPCR verification.3.A logistic regression model was established based on the RT-qPCR data of microRNAs screened from 320 subjects(120 ADHD children and 200 healthy children)summarized in steps 1 and 2.Then the receiver operating characteristic curve(ROC)was used to evaluate the clinical reference value of the logistic regression model for the diagnosis of ADHD.4.From July 2017 to July 2018,362 children were collected as the validation group, including 132 children with ADHD and 230 healthy children.The reliability of the logistic regression model was verified.5.The serum of the follow-up samples in the ADHD children group was collected for RT-qPCR,and the trend of the selected microRNA was observed compared with the newly diagnosed patient data.Results 1.We screened five microRNAs that differed in the serum of ADHD children: miR-4466,miR-4516,miR-6090,miR-4763-3p,miR-4281.Compared with the control group,the expression of miR-4466 was up-regulated,and the expression of the other four miRNAs was down-regulated,and the difference was statistically significant(P<0.001).2.The logistic regression model constructed by the screening group and the training group data was analyzed and analyzed by ROC curve,wherein the AUC was 0.940, the sensitivity was 86.67%,the specificity was 90.00%,and the AUC of the logistic regression model composed of the validated children group data was 0.927.The specificity and specificity were 83.33% and 86.36%,respectively.3.After standard treatment,the expression of miR-4466 in the serum samples of the follow-up children was decreased compared with the first-time children,and the expression levels of miR-4516,miR-6090,miR-4763-3p,and miR-4281 were increased.The difference was statistically significant(P<0.05)ConclusionWe found a group of microRNAs that differed in the serum of children with ADHD and established a logistic regression analysis model to provide a clinical basis for the diagnosis of ADHD.
