Objective: to detect the expression of miR-146 a in peripheral blood of Kazakh patients with rheumatoid arthritis(RA)with damp-heat arthralgia syndrome,and to observe the expression of miR-146 a in peripheral blood of Kazak patients with rheumatoid arthritis(RA)of damp-heat arthralgia syndrome and its correlation with disease activity.Methods: from March 2017 to July 2017,30 Kazakh inpatients with rheumatoid arthritis diagnosed in the Department of Rheumatology and Immunology of the fourth Clinical Medical College of Xinjiang Medical University and classified as damp-heat arthralgia syndrome according to TCM syndrome differentiation were selected.And 30 Kazakh residents who were examined in the fourth Clinical Medical College of Xinjiang Medical University.peripheral blood samples were taken and real-time fluorescence quantitative PCR(FQ-Ct)was used for quantitative statistics.The quantity of the target gene(RQ)= 2-Ct.The expression of miR-146 a in PBMCs of 30 Kazakh patients with damp-heat arthralgia syndrome(RA group)and 30 non-RA patients(non-RA group)was relatively quantified.The number of swollen joints(Swollen Joints:0-28),tenderness joints(Tender Joints:0-28),C-reactive protein(CRP)and erythrocyte sedimentation rate(ESR),)were collected.Analysis of Kazakh dampness by platelet(PLT),anti-cyclic citrullinated peptide antibody(CCP),anti-keratin antibody and rheumatoid factor(RF).Expression of miR-146 a in patients with rheumatoid arthritis with heat arthralgia syndrome and its correlation with disease activity index.Results: the relative expression of miR-146 a in PBMCs of Kazakh patients with rheumatoid arthritis(RA group)(1.708 ±0.375)was significantly higher than that of normal Kazakh residents(non-RA group)(1.025 ±0.225).The difference was statistically significant(t =-8.462,P ? 0.0001).Relative expression of miR-146 a in PBMCs andDAS28 score,course of disease,joint function grade,erythrocyte sedimentation rate,C-reactive protein,platelet,anti-cyclic citrullinated peptide antibody,anti-keratin antibody and rheumatoid factor in RA group.There was no significant difference in the number of samples available(P > 0.05).Conclusion: through this study,the expression of miR-146 a in PBMC of(RA)patients with rheumatoid arthritis of Kazak nationality in Xinjiang is significantly higher than that of normal Kazakh residents in PBMC.There was no correlation between the expression level and DAS28 score,course of disease,joint function grade,erythrocyte sedimentation rate,C-reactive protein,platelet,anti-cyclic citrullinated peptide antibody,anti-keratin antibody and rheumatoid factor.The expression of miR-146 a in PBMC of RA patients was not correlated with the activity of the disease.
