Objectives:To investigate the protective effects of stellate ganglion block on cognitive dysfunction in rats with vascular dementia and its effect on SIRT1/NF-?B signaling pathway,and to investigate the role of SIRT1/NF-?B signaling pathway in hippocampal neuron ischemia-reperfusion injury.Methods:(1)A stellate ganglion block(SGB)model and a vascular dementia(VD)model were established by transection of cervical sympathetic trunk and permanent bilateral common carotid artery occlusion.After VD model and SGB model were established for 8 weeks,the spatial learning and memory ability,serum cytokine production,oxidative stress,and the expressions of SIRT1,I?B?,Bcl-2,NF-?B,Bax,Caspase-3 were detected to evaluate the effects of SGB treatment on rats with vascular dementia.(2)The hippocampal neuronal cell line(HT22)was cultured in vitro.After pretreatment with SRT1720 and EX527,the neuronal ischemia-reperfusion injury was established by treatment with oxygen glucose deprivation for 9h and reperfusion for 24 h.Then,neuron vitality,LDH activity,apoptosis rate and the expressions of SIRT1,I?B?,Bcl-2,NF-?B,Bax,Caspase-3 were detected to evaluate the protective effect of SIRT1 on neuronal ischemia-reperfusion injury.Results:(1)SGB produced significant neuroprotective effects in rats with vascular dementia.SGB treatment attenuated oxidative stress,learning and memory deficits,neuronal apoptosis in the hippocampus.Moreover,SGB enhanced the expressions of SIRT1,I?B? whereas it downregulated serum IL-6,IL-8 and the expression of NF-?B significantly.(2)SRT1720 attenuated neuronal injury induced by OGD.SRT1720 pretreatment reduced LDH release,neuronal apoptosis rate and the expression of NF-?B and caspase-3 whereas it upregulated the expression of SIRT1 and I?B?.While,EX527 pretreatment increased LDH release,neuronal apoptosis rate and the expressions of NF-?B and caspase-3,and decreased the expression of SIRT1 and I?B?.Conclusions:Stellate ganglion block relieves cognitive dysfunction in rats with vascular dementia through SIRT1/NF-?B signaling pathway.Activation of SIRT1/NF-?B signaling pathway may attenuate hippocampal neuronal injury induced by oxygen glucose deprivation and reperfusion.
