Background:Gastric cancer(GC)is one of the common digestive tract malignant tumors in human beings,and it is also one of the major diseases that have seriously threaten human health.The high mortality rate of GC is mainly related to the characteristics of high recurrence and high metastasis after operation.It is shown that engulfment and cell motility proteins play an important role in the process of cell migration.Therefore,it has great clinical significance to explore the molecular mechanism of ELMO protein family in the metastasis of GC,such as early diagnosis,targeted intervention therapy and prognosis judgment.Objective:Exploring the relationship between ELMO3 gene and the development of GC through detecting the expression of ELMO3 in GC tissues.To clarify the role of ELMO3 in the invasion and metastasis of GC,we investigated the effects of changing the expression level of ELMO3 on the biological behaviors of GC cell migration,invasion,cell proliferation in vitro.Methods:1.The expression levels of ELMO3 mRNA and protein in 41 gastric cancer tissues and adjacent normal tissues were detected by RT-qPCR and Western blot respectively.2.In vitro experiments,small RNA interference technology was used to down-regulate the expression of ELMO3 in GC cells SGC7901 and BGC823,which were then used in the following scratch test,chamber migration and invasion test to detect the migration and invasion ability of GC cells.The effect of silencing ELMO3 on cell proliferation and cell cycle was examined through cell activity detection and application of flow cytometry.the localization of ELMO3 in cells and the effect of silencing on the ability of F-actin actin to polymerize were examined by fluorescence microscopy.Results:The results of RT-qPCR and Western blot revealed that the expression of ELMO3 in GC tissues were significantly higher than that in adjacent normal tissues(P < 0.05).Moreover,the expression of ELMO3 in GC tissues with lymph node metastasis were markedly higher than that in GC tissues from patients without lymph node metastasis.Inhibiting the expression of ELMO3 in gastric cancer cells significantly suppressed cell migration,invasion and cell proliferation,as well as the polymerization level of cytoskeleton F-actin,and induced GC cell cycle G1 period arrest.Conclusion:1.The expression of ELMO3 in GC tissue was significantly increased than adjacent normal tissues,and the expression level of ELMO3 was closely related to lymph node metastasis in GC.2.ELMO3 can promote the migration,invasion and the proliferation ability of gastric cancer cells.Moreover,it can also lead to cytoskeleton F-actin polymerization.
