Background:Gestational diabetes mellitus(GDM)can increase the risk of pregnancy and fetus,and there is still a lack of widely recognized early diagnostic markers.To delineate the metabolomic profiling and identify early diagnostic biomarkers in maternal plasma from the pregnant women at 20th week of pregnancy who subsequently developed gestational diabetesmellitus(GDM)using liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(LC-Q-TOF MS).Methods:From the Huzhou Central Hospital and Huzhou Maternal and Child Health Hospital,300 plasma samples of pregnant women with no history of hyperglycemia in the past 20 weeks were collected,and the plasma obtained after centrifugation was stored at-80?.Thirty-four subjects underwent OGTT at week 24,screening 30 pregnant women with GDM for the GDM pregnant group(n=30),and then finding 30 normal-age pregnant women at the same time as the normal pregnancy control group(n=30).LC-Q-TOF MS technique was used to establish non-targeted detection of plasma samples from these two groups of patients,followed by principal component analysis(PCA),partial least squares discriminant analysis(PLS-DA)and orthogonal PLS(OPLS)and other methods are used for analysis.Finally,the VIP(Variable Importance in the Projection)value of the OPLS model(threshold>1)is used,and the p-value of the t-test(threshold value of 0.05)is discover the differential metabolites.The ROC curves were then analyzed for the most significant differences in metabolism,and these indicators were calculated for the sensitivity and specificity of the diagnosis of GDM in order to obtain serum metabolic markers associated with early diagnosis of GDM.Results:Twenty-four out of 975 aligned metabolites were distinguished among GDM plasma and healthy controls.In particular,the level of y-linolenic acid and arachidonic acid was significantly elevated in GDM.Conclusions:The y-linolenic acid and arachidonic acid could be selected as new potent biomarkers for GDM diagnosis and prognosis in early pregnancy;however,they still needto be confirmed from large samples in future.
