Gastric cancer ranks the second leading cause of cancer related age-adjusted morbidity and mortality.The diagnostic rate of early gastric cancer is low,and the therapeutic effect of advanced gastric cancer is unfavorable with the five-year survival rate of 20-30%.LncRNAs play important roles in the biological processes of tumors,including tumor progress,invasion and metastasis.However,the role of lncRNA in the development of gastric cancer and its mechanisms were less reported.The differentially expressed lncRNA and mRNA in gastric cancer tissues were screened by the lncRNA-mRNA expression microarray in our previous study,and series of gastric cancer-related lncRNAs were found.LncRNA-AK023 was up-regulated in gastric cancer tissues,and it was speculated that lncRNA-AK023 might be involved in the development of gastric cancer.However,related researches were rarely reported.Further study on the role of lncRNA-AK023 in gastric carcinogenesis and its potential mechanism will help to elucidate the pathogenesis of gastric cancer and provide new clinical diagnostic and predictive indexes.In this study,the expression of lncRNA-AK023 in gastric cancer tissues was validated by qRT-PCR,and the clinicopathological correlation was analyzed.Using modern,cellular and molecular biology techniques,the expression of IncRNA-AK023 in gastric cancer cell lines was interfered by siRNA and the effects of lncRNA-AK023 knock-down on gastric cancer cell proliferation and migration were studied.Based on bioinformatic analysis,a coding-noncoding gene network network was constructed to predict the potential interactive coding gene of lncRNA-AK023.Methods and materials1.Correlation between lncRNA-AK023 expression and clinicopathological features in gastric cancer1.1.The expression of lncRNA-AK023 in tumor and adjacent paired normal gastric mucosa was determined by quantitative RT-PCR(qRT-PCR).1.2.The clinicopathological characteristics of enrolled patients was collected,including gender,age,tumor size,differentiation,invasive depth,lymphatic metastasis,distant metastasis,the tumor-node-metastasis(TNM)classification and pathological classification.Then,the correlation between lncRNA-AK023 expression and clinicopathological features of patients was analyzed.2.Effect of lncRNA-AK023 on the biological behaviors of gastric cancer cells2.1.By transwell migration assays and scar healing assays,the effect of lncRNA-AK023 knock-down on the migratory ability of gastric cancer cells was studied.2.2.By EDU cell proliferation assays,the effect of lncRNA-AK023 knock-down on the proliferation of gastric cancer cells was studied.3.Analysis of the potential interactive genes of IncRNA-AK023To probe into the possible molecular mechanisms of lncRNA-AK023,coding-noncoding gene network(CNC network)was constructed and the potential coding targets of lncRNA-AK023 was predicted.Results1.LncRNA-AK023 was up-regulated in gastric cancer tissues and associated with lymph node metastasis.2.Knock-down of lncRNA-AK023 could inhibit gastric cancer cell migration and proliferation.3.LncRNA-AK023 was related to a series of coding genes associated with migration and proliferation.
