With the continuous improvement of living standards and the universal popularization of medical treatment,life expectancy has risen substantially and the consequent problem of aging has become more and more serious.According to the China Statistical Yearbook in 2016,over the age of 60 of China's population reached 220 million,accounting for 16.14%of the total population.The problem of osteoporosis and its complications in elderly people is becoming more and more prominent.Worldwide,there is a case of fractures caused by osteoporosis every 3 seconds;the social burden caused by this disease accounts for 0.83%of the total social burden caused by other diseases.Among the people over the age of 50 in China,the prevalence rate of female is 20.7%,and the prevalence rate of male is 14.4%,over the age of 60,the prevalence was significantly higher,more prominent in women.Osteoporotic fracture and its complications have become one of the main causes of death and disability in the elderly,which brings heavy family burden to the patients and severely affects their physical and mental health.Teriparatide,is an active fragment of human parathyroid hormone(PTH)1-34 amino acids from the N-terminal,which can promote bone formation,increase bone density,mainly for the treatment of osteoporosis of postmenopausal women and men over 50 years old.It was approved by the FDA in the United States in 2002 and was produced by Eli Lilly through bioengineering.It was sold mainly in the United States,parts countries of Europe,and began to sell in China after it was imported from France Lilly in 2011.With the development of peptide solid-phase synthesis technology in recent years,the synthesis of teriparatide by chemical methods has become more and more mature.This pape,design and optimize a synthetic route of teriparatide by technology of fragment solid phase synthesis,based on a large number of references and patents.Compared with the reported method,this route simplifies the production steps,shortens the production cycle,increases the production yield,reduces the production cost,and amplifies the production successfully.The production cost of teriparatide can greatly reduce after the implementation of industrial production,thereby lower the price of raw materials,and ultimately to the positive effect on reducing drug prices,relieve the medical burden of patients further.
