Background Myocardial infarction has become the first killer threatening human health.Traditional treatment can only relieve symptoms and can not save the infarcted myocardium.Heart transplantation has many limitations due to lack of donor.Stem cell transplantation is a new effective way to repair cardiac function by stem cell derived cardiomyocytes instead of necrotic myocardium.High-throughput screening techniques have found a large number of differentially expressed long chain noncoding RNAs(LNs)in the differentiation of stem cells into cardiomyocytes,but the function and mechanism of LncRNAs in myocardial differentiation are still unclear.The purpose of this study is to clarify the function and mechanism of non coding RNA LINC01405 in embryonic stem cells and derived cardiomyocytes,and to provide evidence for optimizing the existing myocardial induction methods and improving the efficacy of stem cells in the treatment of myocardial infarction.Results A highly expressed lncRNA-LINC01405 in embryonic stem cells and cardiomyocytes was obtained by high-throughput screening technique.Bioinformatics analysis showed that LINC01405 was a long chain noncoding RNAs located on chromosome 12q.The total length of LINC01405 was 402 bp by rapid amplification of cDNA terminal.The results of real-time quantitative PCR showed that the tissue distribution of LINC01405 was mainly located in the heart tissue,and the high expression cell types were mainly embryonic stem cells,induced pluripotent stem cells and cardiomyocytes.In embryonic stem cells,overexpression of LINC01405 can promote cell proliferation and increase the phosphorylation level of AKT.Interfering with the expression of LINC01405 can inhibit cell proliferation and reduce the phosphorylation level of AKT.In cardiomyocytes,LINC01405 can regulate the expression of MYL2.Cardiomyocytes were injected into SCID.BG immunodeficient myocardial infarction mice and the cardiac function was improved significantly in mice with high expression of LINC01405.Conclusion LINC01405 can affect the proliferation of embryonic stem cells through PI3K/AKT signaling pathway.In myocardial cells,LINC01405 can affect the expression of MYL2.
