ObjectiveTo compare the levels of plasminogen activator inhibitor-1(PAI-1)in patients with venous thromboembolism(VTE)with normal controls.To explore the relationship of three polymorphism of PAI-1 4G/5G with the susceptibility of VTE.MethodsThis is a case-control study.By random sampling,200 healthy subjects and 200 venous thromboembolism patients were diagnosed in the study from January 2016 to May 2018 in Nanjing Gulou Hospital.The clinical data of the two groups were collected,and the levels of plasma PAI-1 and 4G/5G polymorphisms in the promoter region of PAI-14G/5G were determined by enzyme-linked immunosorbent assay(Elisa)and polymerase chain reaction(PCR),respectively.By Hardy-Weinberg balance test,?2 test and excluding confounding factors,the differences of genotype and allele distribution between the two groups were compared,and the correlation between VTE and genotype was analyzed.ResultsHardy-Weinberg equilibrium was observed in both groups.PAI-1 4G/5G genotype was an independent risk factor for VTE.Compared with 5G5G genotype,the risk of VTE was 2.000 times higher in PAI-1-4G4G genotype and 1.507 times higher in 4G/5G genotype.The plasma PAI-1 level in the VTE group was significantly higher than the NC group.The PAI-1 4G/5G gene mutation had an effect on the plasma PAI-1 level,and the 4G mutation allele was related to the increase of the plasma PAI-1 level.ConclusionsIn Chinese population,a significant increase in PAI-1 level is an independent risk factor for VTE,and the PAI-1 4G/5G may be associated with the susceptibility of VTE,and this association may be achieved by the change of PAI-1 concentration.
