Background:Long non-coding RNAs are generally regarded as important regulators in various human diseases.As a IncRNA,HOXD-AS1 has been found to be a tumor promoter in ovarian cancer,glioma and lung cancer.However,the specific effects of HOXD-AS1 on the chemoresistance of cervical cancer cells are still not fully explored.Therefore,HOXD-AS1 may become an important biomarker and target for cervical cancer,and has potential application value in the treatment of cervical cancer.Objective:To investigate the effects of HOXD-AS1 on the cisplatin resistance and migration capacity of cervical cancer cells.Methods:The expression of HOXD-AS1 in cervical cancer tissues and cells was detected by qRT-PCR.The effects of silencing HOXD-AS1 on the proliferation and apoptosis of CaSki-DDP and HeLa-DDP cells were studied by MTT and colony formation assay.Western-blot was used to detect the levels of EMT-related proteins in CaSki-DDP and HeLa-DDP cells after transfection with si-HOXD-AS1.Transwell method was used to detect the migration ability of CaSki-DDP and HeLa-DDP cells after transfection with si-HOXD-AS1.Results:HOXD-AS1 was expressed in cervical cancer tissues,adjacent tissues,cervical cancer cell lines and cisplatin-resistant cervical cancer cells,and HOXD-AS 1 was highly expressed in cervical cancer tissues and cisplatin-resistant cervical cancer cell lines.The difference was statistically significant(P<0.01);Down-regulation of HOXD-AS1 could inhibit the proliferation and promote apoptosis of cisplatin-resistant cells.MTT assay results indicated that down-regulation of HOXD-AS1 decreased IC50 values of CaSki-DDP and HeLa-DDP cells.The difference was statistically significant(P<0.01);Interference with HOXD-AS1 enhanced the expression of epithelial marker protein and inhibited the expression of mesenchymal marker protein in cisplatin-resistant cells.The above differences were statistically significant(P<0.01).Transwell experiments showed that the cell migration and invasion abilities of CaSki-DDP and HeLa-DDP were weakened after transfection of si-HOXD-AS1.Conclusion:HOXD-AS1 is involved in the occurrence and development of cervical cancer,and cisplatin tolerance can be achieved by regulating cell proliferation,apoptosis,invasion and migration.
