Purpose:We adopted the mouse model with IVF-ET to study the long-term vascular condition of IVF-ET produced offspring.Methods:Offspring mice model of IVF-ET were built to compare with the natural born descendant in vascular regulation.Bodyweight and BMI of mice were measured.Serum levels of cardiovascular related cytokines Ang?,NO,ET-1 and PGE2 were tested by ELISA and colorimetric method.Samples of mesenteric vascular were from IVF-ET mice and normal controls.Given the vital functional in regulating vascular contraction which Ca2+signal pathway exhibited,we select 6 representative genes including Calm2,Itpr1,RyR2,RyR3,Cav1.2,Mlck for study.And mRNA expression levels of Calm2,Itpr1,RyR2,RyR3,Cav1.2,Mlck were tested by qRT-PCR.Protein expression levels of Calm2 and Itpr1 were tested by western blot.Results:Mice born under IVF-ET treatment showed raising bodyweights and abnormal BMI after 20th week as well as Serum levels of NO and Ang? were significantly higher than that of the control.Expression levels of Calm2,Itpr1 were up-regulated both in molecular and protein levels(P<0.05),RyR3 was up-regulated in molecular level(P<0.01),Cav 1.2 was up-regulated in molecular level(P>0.05),while RyR2 and Mlck were down-regulated,in molecular level(P<0.01).Conclusion:The IVF-ET progress perturbs expression levels of vascular related genes in the offspring and may contribute to cardiovascular dysfunction.
