Objective To study the anti-inflammatory effect of pigment epithelium-derived factor(PEDF)inhibits macrophage inflammatory response induced by oxidized low density lipoprotein(ox-LDL).Methods RAW264.7 macrophage cells were treated with different concentrations(100,200,400,800 ng/mL)of PEDF for 24 h in vitro.RAW264.7 macrophage cells was pre-intervened with PEDF(100,200,400 ng/mL)for 1 h followed by exposure to 80 mg/L ox-LDL for 24 hours.In addition,RAW264.7 macrophage cells was pre-intervened with 400 ng/mL PEDF for 1 hour followed by exposure to 1 mg/L LPS for 24 hours.Cell viability were detected by CCK-8 and LDH kits,and apoptosis were detected by Annexin V-FITC/PI double staining,respectively.The expression of interleukin-1(IL-1),monocyte chemotactic protein-1(MCP-1),and tumor necrosis factor-α(TNF-α)were analyzed by western blot and ELISA kits.Results PEDF at 100,200 and 400 ng/mL showed no obvious cytotoxic effect on RAW264.7 macrophage cells,while 800 ng/mL PEDF led to significant cytotoxic effect.ox-LDL-induced RAW264.7 macrophage inflammation model was established,and PEDF intervention was performed on the basis of the model.Compared with ox-LDL model group,the cell viability of PEDF intervention group was decreased,while the apoptosis was increased(P<0.05).And PEDF significantly down-regulated the expression of IL-1,MCP-1,and TNF-α protein in a concentration-dependent manner(P<0.05).The LPS group showed the same results as well.PEDF similarly reduced the cell viability of RAW264.7 macrophage cells and down-regulated the expression of IL-1,MCP-1,and TNF-α protein,while increased the apoptosis.Conclusion PEDF can attenuate ox-LDL-induced RAW264.7 macrophage cells inflammatory response and down-regulate ox-LDL-induced IL-1,MCP-1,and TNF-α protein expression in RAW264.7 macrophage cells while promoting macrophage cells apoptosis.It may provide a basis for PEDF to reduce inflammation.
