Objective:To explore the proliferation and invasion in bladder urothelial carcinoma T24 cells by regulating the expression of TET2Materials and Methods: Human bladder urothelial carcinoma T24 cells were used to experimental subjects.The subjects were divided into 7 groups: T24 cells group,blank group,inhibition control group(T24 cells + TET2-NC),TET2-519 inhibition group(T24 cells + TET2-homo-519),TET2-1417 inhibition group(T24 cells + TET2-homo-1417),TET2-3092 inhibition group(T24 cells + TET2-homo-3092),overexpression control group(T24 cells + Blank plasmid),TET2 overexpression group(T24 cells + TET2 overexpression plasmid).Transfection was performed by liposome Lipofectamine 2000,and protein expression of T24 cells in each group was measured by Western blotting.The proliferation of T24 cells in each group was determined by MTT assay.Transwell assay was used to observe the invasive ability of T24 cells in each group.Statistical analysis of experimental data was performed by software of SPSS 22.0.Results:(1)Successful cells transfection was detected by fluorescence microscopy;(2)Western blotting: the protein expression of TET2 was declined in different degrees in TET2-519 inhibition group and TET2-1417 inhibition group and TET2-3092 inhibition group.And the difference between the TET2-519 inhibition group and inhibition group was statistically significant(p<0.05).The protein expression of TET2 in TET2 overexpression group was higher than that in TET2 overexpression control group and blank group(p<0.05).(3)MTT assay: The cell proliferation rate of TET2-519 inhibition group was 17.18%±2%,which was statistically different from the control group(p<0.05).The cell inhibition rate of TET2 overexpression group was 16.82%±8%,which was statistically significant compared with the overexpression control group(p<0.05).(4)Transwell assay: TET2-519 inhibition group had enhanced invasive ability of T24 cells compared with the control group(p<0.05).Compared with the overexpressed control group,the TET2 overexpression group showed a weaker invasive ability of T24 cells(p<0.05).conclusions: Up-regulatiaon the expression of TET2 can inhibit the proliferation and invasion of T24 cells.
