In recent decades,supramolecular hydrogels have been widely studied and applied in various fields.In particular,supramolecular hydrogels formed by self-assembly of small molecules into macromolecules are used in drug delivery,wound healing,metal ion adsorption,functional material design,and biomarker sensing.As the main and effective active ingredient in green tea polyphenols,EGCG has a variety of biological effects such as anti-inflammatory,anti-oxidation,antibacterial,and inhibition of tumor cell proliferation.It is an ideal natural medicine and is widely used in medical and health care.However,its effects on skin wound healing are rarely reported.In this experiment,the preparation of synthetic supramolecular hydrogel-loaded EGCG hopes to better exert its advantages,reduce the inflammation of the wound surface and observe the effect on wound healing with high efficiency and low toxicity.1.Synthesis and Characterization of Supramolecular Hydrogel Precursor:Fmoc-Tyr(tBu)-OH,Fmoc-Phe-OH,2-Naphthylacetic acid is linked to the possession by dehydration condensation on the 2-chlorotrityl chloride resin with a large number of active sites,Nap-F-F-Y was finally cleaved from the resin by a reagent.The collected liquid was purified by lyophilization to obtain a hydrogel precursor powder(Nap-F-F-Y)product.The synthesis of the purified supramolecular hydrogel precursor material was confirmed by high performance HPLC MS HMR techniques.2.Synthesis and characterization of supramolecular hydrogels containing EGCG:different supramolecular waters loaded with EGCG were prepared by using supramolecular hydrogel precursors and EGCG at different drug loading ratios,different temperatures and different gelling agents.The gel was passed through a reverse bottle experiment,transmission electron microscopy and EGCG sustained release experiments to obtain an optimal EGCG supramolecular hydrogel.3.Animal experiment:The incision model and the full-thickness skin removal model were established on the back of KM mice to observe the supramolecular hydrogel(EGCG)in different drug loading ratios.The healing effect,observed by the EGCG-loaded supramolecular hydrogel is beneficial to the healing of mouse wounds.
