Objective:1.To study the possibility of ALDH1 as a potential marker of gastric cancer stem cells and the relationship between dryness of gastric cancer cells.2.To discuss the effect of macrophage(M?)on high expression of ALDH1 gastric cancer cells.Methods:1.Conventional culture and expansion of gastric cancer cell line(SGC-7901);rountine culture of THP-1 monocyte strain,acquisition and culture of M?.2.The ALDH1~highigh gastric cancer cells were obtained from serum-free culture and identified.The dryness-related markers,proliferation,invasion,cloning and other aspects of ALDH1~highigh gastric cancer cell-related dryness were detected to analyze whether ALDH1could be used as a potential marker molecule for gastric cancer stem cells.3.By establishing co-culture system of ALDH1~highigh and ALDH1~lowow gastric cancer cells with M?,detected the ability of survival rate,migration,invasion and cloning of gastric cells with different levels of ALDH1 expression after co-culture,and analyzed the inhibitory effect of M?on gastric cancer cells with different levels of ALDH1 expression.Results:1.Successfully cultured and expanded THP-1 cells induced M?cells by inducing THP-1 monocytes through PMA.2.Compared with ALDH1~lowow gastric cancer cells,ALDH1~highigh gastric cancer cells had higher expression of CD44,Oct-4 and other dry-related markers,and had stronger proliferation,invasion and cloning ability in biological characteristics.3.Successfully established a co-culture system of gastric cancer cell line(SGC-7901)and M?through transwell chamber;it was found that after co-culture,the survival rate,anti-apoptosis ability,invasion,migration and cloning ability of ALDH1~highigh gastric cancer cells were all higher than ALDH1~lowow gastric cancer cells.Conclusion:1.Gastric cancer cells with high ALDH1 expression have stronger stem cell characteristics,and it is possible for ALDH1 as a potential marker of gastric cancer stem cells.2.To some extent,gastric cancer cells with high expression of ALDH1 can antagonize the effect of M?on their biological characteristics.
