Objective: to establish the diagnostic method of hepatitis c virus(HCV)and the identification method of subtype 1b based on serum Raman spectrum.Methods :the serum of each sample was detected by laser Raman spectrometer with a detection wavelength range of 300 to 300 0cm-1.Airpls-pls-svc algorithm was used to process the Raman spectral data and classify the samples.The training set of HCV diagnosis included145 healthy volunteers and 152 HCV patients,and 55 healthy volunteers and 72 HCV patients.The training set for subtype 1b identification was 88 cases of subtype 1b patients and 64 cases of non-subtype 1b patients,while the test set for subtype 1b identification was 51 cases of subtype 1b patients and 15 cases of non-subtype 1b patients.Results :The Raman spectra of serum HCV1 b and non-1b subtypes are similar,and the Raman spectra of 1007 cm-1,1154 cm-1,1519 cm-1 have different peak strengths.Statistical analysis of these Raman spectra showed that the accuracy of type 1b identification was 100%(51/51),and the accuracy of non-1b subtype identification was 93.333 3%(14/15),indicating a case of misdiagnosis.The total accuracy rate was 98.4848%,with a total misdiagnosis of 1case.Conclusion :this study preliminarily proves that Raman spectrum can be applied to HCV diagnosis and HCV1 b subtype identification,which lays a foundation for the development of new HCV diagnosis and typing technology.
