Objective: To investigate the effects of kaempferol on proliferation and apoptosis of glioma U251 cells,and to investigate the anti-glioma activity of kaempferol.Methods: After conventional culture of glioma U251 cells,glutabulil U251 cells were treated with kaempferol at concentrations of 20,40,80,and 160?g/mL,respectively,and phase-inverted microscope was used to observe the concentration of 20,40,80,and 160?g/mL of kaempferol.Morphological changes of glioma U251 cells;CCK-8 method to detect the inhibitory effect of kaempferol on proliferation of human glioma U251 cells,Annexin V-FITC double staining,flow cytometry to detect U251 cell apoptosis,PI single staining Flow cytometry was used to detect the U251 cell cycle.Results:The growth of glioma U251 cells treated with kaempferol at a concentration of 20,40,80,160?g/mL was slow,and the number of cells was significantly reduced.The cells were not tightly contacted,the size was different,and the cell body shrunk and rounded,and the cytoplasm Concentration,nuclear pyknosis,fragmentation,cell transmittance decreased,cell debris suspended in the culture solution,with the increase of drug concentration,cell morphology changes more and more obvious.The results of CCK-8 method showed that kaempferol could effectively inhibit the proliferation of U251 cells,and its effect ofinhibiting cell proliferation was more obvious with the increase of drug concentration and prolonged intervention time.Annexin V-FITC/PI staining showed that kaempferol could induce apoptosis of U251 cells(P<0.01),and the effect of induction of apoptosis was more obvious with the increase of kaempferol concentration;kaempferol effectively induced cell cycle arrest in G2/M Period(P<0.05),with the increase of kaempferol concentration,the effect is more obvious..Conclusion:1.Kaempferol can effectively inhibit the proliferation of glioma U251 cells and promote their apoptosis.2.Kaempferol effectively induced the cell cycle arrest of glioma U251 cells in G2/M phase.
