Objective: To study the biological characteristics of polyploid cancer cells and its role in malignant phenotype of esophageal squamous cell carcinoma.Methods: The formation of polyploid cancer cells in esophageal squamous cell carcinoma cells was induced by cobalt chloride and paclitaxel.The esophageal squamous cell carcinoma cells and the polyploid cancer cells formed by them were respectively made into cell blocks,immunocytochemical staining was performed to detect the differences in stem cell characteristics,proliferation and epithelial mesenchymal transformation between them.The difference in invasion ability between the two was detected by Transwell invasion assay,and the signal pathway of EGF/EGFR/AKT was detected by Western Blot.Results: Compared with common esophageal squamous cell carcinoma cells,polyploid cancer cells formed by esophageal squamous carcinoma cells showed stronger expression of stem cell characteristics and proliferative markers by immunocytochemical staining,weaker expression of epithelial markers and stronger expression of mesenchymal markers.Transwell invasion assay showed that polyploid cancer cells were more aggressive(P<0.001).Western Blot analysis revealed the activation of AKT signaling pathway in polyploid tumor cells.Conclusion: In this study,it was found that polyploid cancer cells hadcharacteristics of tumor stem cells,enhanced proliferation,and epithelial mesenchymal transformation.Polyploid tumor cells could invade through 8 ?m pore size,and their AKT signaling pathway was activated.In summary,polyploid cancer cells contribute to the malignant phenotype of esophageal squamous cell carcinoma.
