Objective:To explore the effect of PAR-2(protease-activated receptors-2)on the epithelial barrier function and the expression of TJs(tight junctions)in nasal mucosa of AR(allergic rhinitis)patients and its underlying pathogenesis and provide new clues for the treatment and prevention of AR.Method:We detected TJs and PAR-2 in nasal mucosas from AR patients and nomal control group,using IHC,RT-qPCR,and Western blot.We analyzed the correlation between PAR-2 and TJs expression in AR.We also evaluated the influence of PAR-2 expression on TJs in human primarily cultured nasal epithelial cells(HNEpCs).Derp-1 was used to stimulate HNEpCs.PAR-2 antagonist and agonist were used to stimulate cells for 24 hours,respectively.The expression of PAR-2/TJs was detected by q-PCR.Fluorescein Isothiocyanate-dextran(FD4)detection was used as an indicator of epithelial barrier function.Result:We observed significantly elevated PAR-2 production both in AR group and HNEpCs after Der p1 stimulation.The expression of Claudin-1,-3,-7,-12?JAM-A?Occludin?ZO-1 decreased significantly in AR group.After correlation analysis,ZO-1,Claudin and PAR-2 expression were negatively correlated(p < 0.05).Conclusion:These findings demonstrate that the PAR-2 induction is involved in epithelial barrier dysfunction and TJs expression decreased,suggesting that PAR-2 may play an important role in the pathogenesis of AR. |
