Objective:This study aimed to explore the effects of Klotho in human tongue cancer cells and underlying mechanisms.Methods:1.Western Blot has been applied to examine the protein expression level of Klotho,?-catenin,c-Myc,Cyclin D1,Caspase-3 and so on.2.MTT assay and colony formation assay have been used to examine the proliferation ability of cells.3.The cell apoptotic rate were measured by using flow cytometry assay.Results:1.In the MTT assay,cell proliferative ability in the experimental group were lower.2.There were fewer colonies in the experimental group.3.Compared with control group,the expression of ?-catenin,c-Myc and Cyclin D1 were decreased,while Caspase-3 was increased.4.Klotho markedly increased the cell apoptotic rate.Conclusion:Klotho can inhibit the proliferation ability and induce apoptosis of Cal27 cells.These phenomena are partly because Klotho can inhibit Wnt/?-catenin signaling pathways.
