Objective:By applying ~1H-NMR metabolomics research technique and adopting diversified comprehensive analysis techniques and methods,the blood metabolite spectra of 36 patients with renal Yin deficiency and renal Yang deficiency of menopausal syndrome were compared and analyzed,in order to find specific metabolic markers of the syndromes of renal Yin deficiency and renal Yang deficiency and provide theoretical basis for the microscopic differentiation of TCM syndromes of menopausal syndrome.Methods:Reference to disease and standard syndromes,kidney Yin deficiency syndrome in postmenopausal syndrome patients,all the 36 patients with kidney Yang deficiency syndrome,and to detect its blood metabolites,obtain metabolites spectrum,by means of chemometrics for these information extraction,processing,and get the score chart and the load diagram,the comparative analysis between patients with kidney Yin deficiency syndrome,kidney Yang deficiency metabolic product spectrum characteristics,specific metabolic markers,finding potential syndromes.Results:(1)The biomarkers of renal Yin deficiency in menopausal syndrome were isoleucine,lactic acid,glutamic acid,pyruvate,n-acetylglycoprotein,acetic acid and TMAO.The contents of isoleucine,lactic acid,glutamic acid,pyruvate and n-acetylglycoprotein were significantly down-regulated(P<0.05 or P<0.01),while the contents of acetic acid and TMAO were significantly increased(P<0.05 or P<0.01).(2)The biomarkers of renal Yang deficiency syndrome of menopausal syndrome are:lactic acid,glutamic acid,choline,creatine,acetic acid,n-acetylglycoprotein.The contents of lactic acid,glutamic acid,choline,creatine and n-acetylglycoprotein are significantly decreased(P<0.05 or P<0.01),while the contents of acetic acid are significantly increased(P<0.01).(3)The metabolic biomarkers for the differences between the two syndromes of renal Yin deficiency and renal Yang deficiency in menopausal syndrome may be-glucose,choline,creatine,TMAO,glycerol phosphocholine,n-acetylglycoprotein and-glucose(AUC>0.85),but the differences need to be confirmed by further studies.(4)Renal Yin deficiency syndrome of menopausal syndrome is mainly related to pyruvate metabolism,glycolysis and gluconeogenesis,alanine,aspartic acid and glutamic acid metabolism,butyric acid metabolism,glutamic acid and glutamine metabolism,TCA cycle and other pathways.(5)Renal Yang deficiency syndrome of menopausal syndrome is mainly related to pyruvate metabolism,alanine,aspartic acid and glutamic acid metabolism,glutamine and glutamic acid metabolism,arginine and proline metabolism,sulfur metabolism and aminoacyl-tRNA biosynthesis pathway.Conclusion:Menopause syndrome in patients with kidney Yang deficiency and kidney Yin deficiency syndrome serum metabolomics markers dissimilarity,beta glucose,choline,creatine,TMAO,glycerol phosphate choline,N-acetyl glycoprotein,alpha glucose(AUC>0.85)has high diagnostic value as well as the ability to predict,it may be associated with menopause differences in kidney Yang deficiency and kidney Yin deficiency syndrome subjects both metabolic biomarkers.
