Objective:To investigate the effects of dexmedetomidine combination with Mild Hypothermy on cognitive function in Sepsis associated encephalopathy rats,by comparing the effects of dexmedetomidine combination with Mild Hypothermy with only dexmedetomidine or Mild Hypothermy,and assess the possible mechanism,providing an effective clinical and theoretical basis for the prevention of Sepsis associated encephalopathy.Methods:Slected 80 male SD rats,weighing 300-350g,were equally and randomly divided into 5 groups using a random number table:sham group?group S?,sepsis group?group CLP?,dexmedetomidine group?group D?,Mild Hypothermy group?group H?and dexmedetomidine combination with Mild Hypothermy group?group DH?.Sepsis model was reproduced by cecum ligation perforation in group CLP,D,H and DH.The temporalis of the rats in both groups S,CLP and D was kept at a temperature of 37.5?for 30 min and that of the rats in both groups H and DH at 35?for the same period of time before CLP model.When temperature stability then intraperitoneal injection of 100 ug/kg dexmedetomidine in group D and DH.Four rats were chosen from each group selected at the time before CLP?T0?and at 4,5 and 6 days after CLP?T1-T3?respectively underwent Morris water-maze test for assessment of the cognitive function and neurologic deficit score.The hippocampal was isolated,and the active of SOD?MDA in hippocampal were detected by chemical colorimetry,And hippocampal samples were taken for determination of the levels of TNF-a,IL-6 and IL-10 by Elisa.Results:?1?Compared with group S,the escape latency was prolonged,the frequency of crossing the original platform was decreased,and the neurologic deficit scores were increased in other groups?P<0.05?.Compared with group CLP,the escape latency was shortened,the frequency of crossing the original platform was increased,and the neurologic deficit scores were decreased in group D,H and DH?P<0.05?.Compared with group DH,the escape latency was prolonged,,the frequency of crossing the original platform was decreased,and the neurologic deficit scores were increased in group D and H?P<0.05?.?2?Compared with group S,the activition of SOD in hippocampal was decreased,and activition of MDA was increased in other groups?P<0.05?.Compared with group CLP,the activition of SOD in hippocampal was increased,and activition of MDA was decreased group D,H and DH?P<0.05?.Compared with group DH,the activition of SOD in hippocampal was decreased,and activition of MDA was increased group D and H?P<0.05?.?3?Compared with group S,the hippocampal TNF-a,IL-6 and IL-10 were increased in other groups?P<0.05?.Compared with group CLP,the hippocampal TNF-a and IL-6 were decreased,hippocampal IL-10 was increased in group D,H and DH?P<0.05?.Compared with group DH,the hippocampal TNF-a and IL-6 were increased,hippocampal IL-10 was decreased in group D and H?P<0.05?.Conclusion:Dexmedetomidine combination with Mild Hypothermy can improve cognitive function in Sepsis associated encephalopathy rats,and is better than the effect of dexmedetomidine alone or mild hypothermy alone,and inhibition of hippocampal Oxidative stress injury and Inflammatory reaction may be involved in the underlying mechanism.
