Study On The Intervention Mechanism Of Endoplasmic Reticulum Stress In AS Habbits After PTCA With Huotan Jiedu Tongluo Decoction

Objective: To investigate the effect of huotan jiedu tongluo decoction on rabbit carotid artery restenosis model after PTCA,and whether it can inhibit IRE1α/XBP1 signaling pathway improves intimal hyperplasia.Method: 30 rabbits were divided into 5 groups,6 rabbits in each group.Sham operation group,model group,TCM group(hutan detoxification tongluo group),TCM high-dose group,western medicine group(atorvastatin group).The rabbits in the 5 groups were given ordinary diet for 7 days after adaptive feeding.Except the sham group,the other 4 groups were given 2% l-methionine for 14 days before PTCA.Rabbit carotid atherosclerosis model was established by common carotid artery balloon injury,and samples were obtained after continuous gavage for 28 days on the second day after surgery.The pathological morphology of the injured carotid artery was observed by HE staining.The serum HCY concentration of the rabbit was detected by ELISA,the mRNA expression of IRE1,XBP1 in the rabbit carotid artery was detected by qPCR,and the protein expression of IRE1,XBP1 in the rabbit carotid artery was detected by western-blot.Result:Compared with the sham operation group,the vascular endodermis in the model group was significantly thickened and the lumen was narrowed.Epithelial hyperplasia with inflammatory cell infiltration.Compared with the model group,the intimal hyperplasia and the degree of lumen stenosis were reduced in the traditional Chinese medicine group and the western medicine group.The serum HCY concentration of each group was lower than that of the model group,The concentration of TCM group was the lowest.The qpcr results showed that compared with the sham operation group,the mRNA content of IRE1 and XBP1 in the model group was significantly increased,and compared with the model group,the HTJDTL group was significantly increased after intervention with atorvastatin group,its mRNA content was significantly reduced.Western blot results showed that the expression of histone in the model group was significantly increased compared with that in the sham operation group compared with the model group,the protein expression of HTJDTL and atorvastatin significantly decreased after intervention.Conclusion: 1.Huatan jiedu tongluo can significantly reduce the area of carotid artery endothelial hyperplasia and improve vascular inflammation.Alleviate carotid artery intimal stenosis,and the effect is better than western medicine group.2.Hutan jiedu tongluo can reduce the content of HCY in rabbit serum,which is helpful for hyperhomocysteinemia It’s inhibitory.3.The expression of IRE1,XBP1 protein and mRNA can be down-regulated by hutan detoxification tongluo.instructions Hutan jiedu tongluo fang may inhibit the hyperplasia of arterial intima through IRE1-xbp1 pathway.
