ObjectiveTo investigate the expression of sphingosine-1-phosphate transporter 2(Spinster homolog 2,SPNS2)in colorectal cancer and its clinical significance,and to determine the relationship between SPNS2 and prognosis of patients with colorectal cancer.MethodsImmunohistochemistry was used to detect the expression of SPNS2 protein in 133pairs of colorectal cancer and adjacent normal tissue sections.Quantitative Real-time PCR(qRT-PCR)was used to detect the expression of SPNS2 in 29 pairs of colorectal cancer and adjacent normal tissues.The survival curve of the relationship between the difference of SPNS2 expression and the overall survival rate(OS)of patients with colorectal cancer was drawn.?~2 test was used to analyze the relationship between the expression of SPNS2 and the clinicopathological features(age,sex,differentiation,number of lymph node metastases(pN),tumor size and depth of invasion(Tx)of patients with colorectal cancer.Kaplan-Merier survival analysis was used to analyze the relationship between gender,age,number of lymph node metastases(pN),tumor size,SPNS2 expression,distant metastasis(M),depth of invasion(Tx),differentiation and prognosis of patients with colorectal cancer.Log-rank was used to compare the differences between groups.Cox multivariate analysis was used to analyze the independent influencing factors of prognosis of patients with colorectal cancer.ResultsThe results of immunohistochemistry showed that the expression of SPNS2 in colorectal cancer tissues was higher than that in adjacent normal tissues(81.2%VS22.6%,?~2=69.136,P<0.001).RT-q PCR results showed that the expression of SPNS2 in colorectal cancer tissues was significantly higher than that in corresponding adjacent normal tissues(t=3.974,P<0.001).Overexpression of SPNS2 protein was closely related to the degree of differentiation,number of lymph node metastasis(pN),tumor size and depth of invasion(Tx),with statistical significance(P<0.001).The survival time of patients with negative SPNS2 expression was significantly longer than that of patients with positive SPNS2 expression(?~2=13.080,P<0.001).Univariate analysis showed that the number of lymph node metastasis(pN),tumor size,abnormal expression of SPNS2,distant metastasis(M),depth of invasion(Tx),and degree of differentiation were correlated with prognosis of patients with colorectal cancer(P<0.05),while age and gender of patients were not significantly correlated with prognosis.Cox multivariate analysis showed that the number of lymph node metastasis(pN),abnormal expression of SPNS2,distant metastasis(M),and depth of invasion(Tx)were independent prognostic factors for colorectal cancer(P<0.05).ConclusionsThe positive expression of SPNS2 in colorectal cancer is related to the occurrence and development of colorectal cancer.SPNS2 can be used as a new marker of colorectal cancer.It can be used to evaluate the prognosis of patients,monitor the invasion and metastasis of tumors after operation,and serve as a new target for diagnosis and treatment.
