Expression And Clinical Significance Of MiRNA-150 In Peripheral Blood Mononuclear Cells Of Primary Gouty Arthritis

Objective: To investigate the potential role of miR-150 involved in the pathogenesis of primary gouty arthritis.Methods:(1)The peripheral blood and clinical data of 98 gout patients(GA group),39 healthy controls(HC group)and 40 osteoarthritis patients(OA group)were collected,and the 98 gout patients were divided into acute gout patients(AG group,n=34),interval gout patients(IG group,n=34)and chronic gout patients(CG group,n=30);(2)Taq-Man polymerase chain reaction(Taq-Man PCR)was used to detect the expression of miR-150 in peripheral blood mononuclear cells(PBMCs).Real-time quantitative polymerase chain reaction(RT-qPCR)was used to detect the expressions of MyD88,P2X7 R and SOCS1 mRNA in PBMCs;(3)Western Blot was used to detect the protein levels of MyD88,P2X7 R and SOCS1 in PBMCs.(4)Statistical analysis was performed using SPSS 23.0 software.Data were displayed as median(quartile range)or mean ± standard deviation.One-Way ANOVA,independent-samples T test and Spearman's correlations analysis were used for statistical analysis.P<0.05 is defined as statistical significance.Results:(1)The age of OA group was significantly higher than GA group and HC group(p<0.05),the levels of ESR and WBC in GA group were significantly higher than HC group(p<0.05),and the level of WBC in GA group was higher than OA group(p<0.05).(2)There was no significant difference in age among AG,IG,CG and HC groups(p>0.05).The levels of WBC?GR?hsCRP and IBIL in AG and CG group were higher than those in HC group(p<0.05).The levels of ALT,TG,VLDLC,LDLC,Crea and UA in different phases of gout were higher than those in HC group,the level of HDLC was lower than that in HC group;(3)The expressions of miR-150 in GA group and OA group were lower than those in HC group,but the differences were not statistically significant(P>0.05).(4)MiR-150 expression in the AG group was lower than that in the IG,CG and HC groups(P<0.05),Meanwhile the expressions of miR-150 in the IG and CG groups were lower than that in the HC group(P>0.05);(5)The expression of miR-150 in AG group was negatively correlated with WBC,GR,RBC,HGB,DBIL and IBIL(p<0.05).The expression of miR-150 in CG group was positively correlated with GR,LY,Mo and DBIL(p<0.05),and negatively correlated with HGB and PLT(p<0.05).The expression of miR-150 in HC group was positively correlated with hsCRP(p<0.05).No correlation was observed between the expression level of miR-150 and various clinical data in IG group,and in OA group as well(p>0.05);(6)MyD88 mRNA and protein levels in the GA group and OAgroup were higher than those in the HC group(P<0.05),and the expression of MyD88 mRNA in the GA group was higher than that in the HC group(P<0.05).When the GA group was divided into three groups,the levels of MyD88 mRNA and protein in different stages of gout were all higher than those in the HC group(P<0.05,respectively).The levels of MyD88 mRNA and protein in AG group were the highest among those groups(P<0.05).The MyD88 mRNA expression in AG group was negatively correlated with miR-150(p<0.05)and positively correlated with ESR,WBC and GR(p<0.05).(7)The expressions of P2X7 R mRNA in GA and OA groups were higher than that in HC group(P<0.05).When the GA group was divided into three groups,the P2X7 R mRNA expression in AG,IG,CG and OA group were all higher than that in HC group(P<0.05).The expression of P2X7 R mRNA in AG group was higher than that in IG,CG and OA group(P<0.05).The P2X7 R protein level of PBMC in each group were not detected successfully;The P2X7 R mRNA expression in OA group was negatively correlated with miR-150(p<0.05);(8)No obvious differences of SOCSI mRNA and protein expression levels were observed among these groups(P>0.05),while the expression of SOCS1 mRNA in different stages was not correlated with miR-150(p >0.05).Conclusion:(1)The elevated levels of WBC,GR and hsCRP in AG group indicated that WBC,GR and hsCRP may have the potential to reflect the severity of gout inflammation.The raise of ALT,IDIL,TG,HDLC,LDLC,apoB1,Crea and UA levels in different stages of gout suggested that the injury of liver cells,disorder of blood lipid metabolism and renal dysfunction may exist in patients with gout.Meanwhile,it is important to maintain the goal-directed therapy of reducing uric acid in intermittent and chronic gout;(2)The miR-150 expression in GA group and OA group decrease,suggesting that miR-150 may play a role in the pathogenesis of gout and osteoarthritis;(3)Decreased protein level in acute gout stage revealed that miR-150 may be related to the severity of inflammation;(4)The association between miR-150 expression with clinical data suggested that miR-150 may be related to the inflammatory process of gouty pathogenesis through multiple pathways;(5)The trend of MyD88 mRNA and protein changes among these groups and the negative correlation between the MyD88 mRNA expression and miR-150 in AG group revealed that miR-150 may be involved in the occurrence and development of gouty inflammation by regulating MyD88 expression.The expression of MyD88 mRNA and protein in GA group were higher than that in OA group,which may be related to the more intense inflammation of gout arthritis compared with osteoarthritis;(6)Changes in the expression of P2X7 R mRNA among groups and the negative correlation between the P2X7 R mRNA and miR-150 expression in OA group suggested that P2X7 R might be associated to the occurrence of gout and accept the regulation from miR-150;(7)There was no significant difference of SOCS1 mRNA or protein level was observed among groups,moreover,the SOCS1 mRNA expression in every stage was not correlated with miR-150,suggesting that SOCS1 expression may not be related to gout and miR-150 expression.
