Background In recent years,studies have shown that gut microbiota has played a key role in steady state and the host immune response,psoriasis for polygenic inheritance of immune-mediated skin disease,a growing body of research suggests that gut microbiota may pathogenic role in the pathogenesis of psoriasis,psoriasis and the composition of gut microbes groups but still not reached a consensus.Objective In order to understand the relationship between psoriasis and intestinal microorganisms,we observed the microflora structure and mutual differences between psoriasis patients and normal healthy people through 16 S sequencing,providing a favorable basis for insight into the pathogenesis of psoriasis and the development of new treatment.Methods 20 patients with chronic plaque psoriasis and their corresponding 20 healthy controls were selected to collect fecal samples of the subjects for DNA extraction,PCR amplification of target fragments,library construction,on-board sequencing,biological information analysis and other steps.Results 1.84746 reads were tested from each sample on average,and 80155 effective data were obtained on average through quality control.The quality control efficiency was up to 94.63%.The sequences were clustered into OTUs with 97% consistency,and a total of 1362 OTUs were obtained.We found that at the phylum level,the top 10 species with relative abundance included:Firmicutes,Bacteroidetes,Proteobacteria, Actinobacteria,Tenericutes,Verrucomicrobia,Euryarchaeota,Fusobacteria,Melainabacteria and unidentified_Bacteria.On the genus level,Bacteroides,Megamonas,unidentified_Enterobacteriaceae,Faecalibacterium,Bifidobacterium,unidentified_Lachnospiraceae,Blautia,Holdemanella,Streptococcus and unide were applied Ntified_Prevotellaceae;3.There was no significant difference in the bacterial community richness and diversity between the psoriasis group and the healthy control group,but the two groups were significantly different in the microbial community composition.About the phylum level,Euryarchaeota,Cyanobacteria,Chloroflexi and Rokubacteria are found in relatively high abundance in patients with psoriasis.On the generic level,Gilliamella(the genus of chlamydia),Methanobrevibacter,Terrisporobacter,Bartonella(the genus of Bartonella),Catenibacterium,Stenotrophomonas(oligotrophomonas),Methanosphaera(the genus of chlamydia),Oscillibacter,Peptoclostridium were relatively abundant in psoriasis patients.Porphyromonas(Porphyromonas)and Stoquefichus were less abundant.By linear discriminant analysis,it was found that Negativicutes,Lactobacillales,Romboutsia,Bacilli,Selenomonadales,Megamonas and other 7 intestinal biomarkers were identified.Conclusion This study identified the microflora structure of patients with psoriasis and healthy control population,and determined the significant difference between intestinal microflora of silver chip patients and healthy population,providing a new and comprehensive approach to explore the symbiotic relationship between intestinal microflora and psoriasis.
