| Objective This study investigated the expression of miR-125a-5p in the serum of patients with nonsmall cell lung cancer,to explore whether miR-125a-5p can be used as a miRNA marker of peripheral blood serum of patients with non-small cell lung cancer.The relationship between the expression of miR-125a-5p and its clinicopathological characteristics was analyzed,providing a new direction for the early diagnosis and prognosis of non-small cell lung cancer.Methods 1.From February 2017 to February 2018,44 newly diagnosed patients with non-small cell lung cancer who were admitted to the respiratory department of Chaohu Hospital of Anhui Medical University were collected.During the same period,20 healthy subjects from the health management center were selected.Patients who met the enrollment criteria and agreed to participate in the study and healthy subjects were included in the study.2.The expression level of miR-125a-5p in peripheral blood of 44 patients with non-small cell lung cancer and 20 healthy volunteers was detected by real-time quantitative PCR,and the difference of miR-125a-5p expression between the two groups was compared.The differences in miR-125a-5p expression between 20 healthy controls,9 patients with stage I/II NSCLC,and 35 patients with stage III/IV NSCLC were further analyzed.3.The expression levels of CEA and cyfra21-1 in peripheral blood of 44 patients with non-small cell lung cancer and 20 healthy volunteers were detected by electrochemical luminescence,and the expression levels of CEA and cyfra21-1 were compared between the two groups.4.To analyze the correlation between the expression level of serum miR-125a-5p and the serum tumor markers and clinicopathological characteristics of non-small cell lung cancer.5.The area under the ROC curve of serum miR-125a-5p and its combination with CEA and cyfra21-1 detection for the diagnosis of non-small cell lung cancer was calculated respectively.6.Statistical analysis: SPSS19.0 statistical software was used to analyze and process the data,including kruskal-wallis test,non-parametric test and Spearman rank correlation analysis.Results 1.The relative expression level of miR-125a-5p in peripheral blood serum of the nonsmall cell lung cancer group was 0.025(0.017,0.051),lower than that of the healthy group(0.050,0.034,0.117),and the difference was statistically significant(P<0.05).The relative expression level of miR-125a-5p was significantly different between healthy control group,stage I/II NSCLC group and stage III/IV NSCLC group(P<0.001).The relative expression level of miR-125a-5p in group III/IV patients was 0.019(0.012,0.029),which was lower than that in group I/II 0.064(0.055,0.085).The difference was statistically significant(P<0.05).The area under the ROC curve of miR-125a-5p for the diagnosis of non-small cell lung cancer was 0.804.The area under the ROC curve of miR-125a-5p combined with CEA and cyfra21-1 detection for the diagnosis of non-small cell lung cancer was 0.998.2.The expression level of CEA in NSCLC group was 4.370(2.603,11.573)ng/mL,which was higher than that in healthy group1.385(0.875,2.060)ng/mL,and the difference was statistically significant(P<0.001).The expression level of cyfra21-1 in NSCLC group was 5.460(0.260,1.638)ng/mL,which was higher than that in healthy group(0.820(0.260,1.638)ng/mL,the difference was statistically significant(P<0.001).3.The correlation between the relative expression level of miR-125a-5p in peripheral blood of NSCLC group and its clinicopathological characteristics was analyzed,and no significant difference was found in the expression level of miR-125a-5p between gender,smoking,age and tissue classification(P>0.05).4.Spearman rank correlation analysis showed that the relative expression level of miR-125a-5p in peripheral blood of NSCLC was negatively correlated with clinical TNM stage(r=-0.688,P<0.05),but was not correlated with the expression level of serum tumor markers CEA and cyfra21-1(P>0.05).Conclusion 1.miR-125a-5p is down-regulated in peripheral blood serum of NSCLC,and the expression level of miR-125a-5p is negatively correlated with its clinical TNM stage,suggesting that miR-125a-5p may be a biological indicator for diagnosis of NSCLC.2.The combined detection of miR-125a-5p with CEA and CYFRA21-1 in peripheral blood of non-small cell lung cancer can improve the diagnostic value of NSCLC. |
