Objective: To explore the effect of HBV infection on hepcidin expression and the preliminary study of hepcidin anti-HBV activity.Methods: Serum samples were collected from 71 patients with CHB(CHB group)and24 healthy subjects(control group).Serum hepcidin(Hepc)levels were measured by ELISA.Detection of serum iron(SI),ferritin(SF)and transferrin(TRf)levels using a fully automated protein analyzer.Detection of serum IL-6 levels using an electro-chemiluminescence automated immunoassay analyzer.According to the results of recent patients with pathological inflammatory activity(G),the CHB components were compared with Hepc,SI,SF,and TRf in the mild inflammatory group(G1-G2),the moderate group(G3),and the severe group(G4).The levels of HBV-DNA,HBs Ag and HBe Ag in CHB group were measured,and the correlation between hepcidin level and virological index and liver function biochemical index was analyzed.The Huh7 cell model of HBV infection was constructed,and the expression of hepcidin in hepatocytes was detected by RT-q PCR and Western blot.HepG2 cells transfected with pcDNA1.3 and Huh7 cells transfected with pSM2 were treated with hepcidin.The levels of HBV-DNA and HBe Ag in the culture supernatant between the hepcidin-treated and untreated groups were detected and analyzed at different time points.difference.Results: Compared with the control group,SI,SF,IL-6,alanine aminotransferase(ALT),serum aspartate aminotransferase(AST),and serum total bilirubin(TBIL)were significantly elevated in the CHB group.Hepcidin,TRf and soluble transferrin receptor(s Tf R)and albumin(ALB)were significantly decreased(P<0.05).Serum hepcidin levels in patients with CHB were significantly higher than HBV-DNA(r=-0.467,P<0.05),HBs Ag(log10)(r=-0.567,P<0.05),and HBe Ag(r=-0.434,P<0.05).There was a significant positive correlation between serum ALB and TBIL levels(P<0.05),and no significant correlation with ALT and AST levels(P>0.05).After transfected cells for 24 h,the relative level of Hepc m RNA was(5.21±0.43),and the control group(0.73±0.14)was significantly increased(P<0.05).At 48 h after transfection,the cell Hepc m RNA level was(8.45 ± 0.61).),compared with the blank control(1.16 ± 0.17)significantly increased(P<0.05);after 48 h of plasmid transfection,the relative expression of Hepc protein was(0.78±0.08),compared with the blank control(0.41±0.02).Significantly elevated(P<0.05)Compared with the untreated group,the pc DNA1.3 transfected Hep G2 cells in the hepcidin treatment group showed a decrease in HBV DNA levels at 12 h,and a significant decrease in HBV DNA levels at 48 h(P<0.05).p SM2 transfected Huh7 cells with iron regulation The HBV DNA level decreased at 24 h and the HBV DNA level decreased significantly at 48h(P<0.05).Compared with the untreated group,the pc DNA1.3 transfected Hep G2 cell hepcidin treatment group showed a decrease in HBe Ag level at 24 h,and a significant decrease in HBe Ag at 48h(P<0.05).p SM2 transfected Huh7 cell hepcidin treatment group HBe Ag levels decreased at 24 h and HBe Ag decreased significantly at 48 h(P < 0.05).Conclusions: Detection of hepcidin and iron metabolism-related indicators might help to assess the progress of chronic hepatitis B,and an appropriate administration of hepcidin or its corresponding endogenous activators needs clinical trials,and initially proved that hepcidin has certain inhibition of HBV replication.
