Objective: To observe the effects of genistein(GEN)on the proliferation of normal thyroid cell line and thyroid cancer cell lines,and the effects of GEN combined with UV on proliferation of normal thyroid cell line Nthy-ori-3-1,thyroid papillary carcinoma cell line TPC-1 and human undifferentiated thyroid cancer cell line 8305 C.To explore the possibility of the application of genistein in the treatment of thyroid cancer.Methods: Nthy-ori-3-1,TPC-1,8305 C cells were cultured routinely.Three kinds of cells were divided into two groups: drug-treated group and drug-UV irradiation group.Different doses of genistein were added to the culture medium of the two groups of cells,and the cells of drug-UV irradiation group were irradiated with ultraviolet rays at the wavelength of 254 nm.The3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT)assay was used to observe the effect of drug alone treatment group and drug combined ultraviolet ray group on cell proliferation.Results:The results of MTT showed that the proliferation of Nthy-ori-3,TPC-1 and 8305 C cells were inhibited by genistein in the drug-treated group,and the degree of inhibition increased with the increase of the drug concentration.The proliferation of the three cell lines in combination with ultraviolet radiation was inhibited,and the inhibition degree was greater than that of single drug treatment.Conclusion:Genistein can inhibit the proliferation of thyroid cell line Nthy-ori-3 and TPC-1,undifferentiated thyroid cancer cell line 8305 C,and the inhibitory effect is stronger when combined with ultraviolet radiation.
