Objective to investigate the relationship between NLRP3 and related factors and the progression of aortic stenosis by detecting the protein levels of peripheral blood NLRP3 and related factors IL-1? and IL-18 in patients with aortic stenosis.Methods 60 patients with moderate or more aortic stenosis in the Cardiovascular Medicine Clinic and Heart Research Center,First Affiliated Hospital of Guangxi Medical University from April 2018 to April 2019 were selected as observation group.They were divided according to the medical history,cardiac color doppler ultrasound and surgery such as seen in the aortic valve morphology.Among them,there were 30 cases of calcific aortic stenosis(CAS),30 cases of rheumatic aortic stenosis(RAS).45 inpatients without valvelar heart disease examined by cardiac color doppler ultrasound were collected as the control group during the same period.The levels of NLRP3,IL-1? and IL-18 proteins in peripheral blood of each group were determined by enzyme-linked immunosorbent assay(ELISA),and the levels of hypersensitive c reactive protein,white blood cell count and amino terminal b-type natriureticpeptide and so on were detected by blood biochemistry.The above data were analyzed by SPSS20.0 statistical software.Results 1.The levels of NLRP3,IL-1? and IL-18 in the peripheral blood of patients with aortic stenosis were significacntly higher than those in the control group(P<0.05),and the levels of NLRP3,IL-1? and IL-18 in the RAS group were all higher than those in the CAS group(P<0.05).2.The levels of peripheral blood NLRP3 was positively correlated with IL-1?,IL-18,hs-CRP,WBC,NT-pro BNP and was negatively correlated with LVEF in patients with aortic stenosis.Conclusion 1.Peripheral blood NLRP3 and its downstream factors in patients with aortic stenosis were higher than that in normal people,and peripheral blood NLRP3 was positively correlated with IL-1?,IL-18,hs-CRP,WBC in patients with AS,suggesting that inflammation related to NLRP3 inflammatory corpuscle may be involved in the process of aortic stenosis.2.The expression level of NLRP3 in patients with aortic stenosis was positively correlated with NT-pro BNP and was negatively correlated with LVEF,which may cause inflammatory response through the downstream inflammatory cytokines of NLRP3 inflammatory bodies,leading to changes in cardiac valve structure,stenosis and decreased left ventricular ejection function.
