ObjectivesTo detect the expression of ASH2L and NF-?B signaling pathway activating protein molecules P65 and I?B?in superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia and gastric cancer.And to investigate the relationship between the expression of ASH2L and the activation of NF-?B signaling pathway in different gastric mucosal lesions associated with helicobacter pylori infection.Methods1 Tissue specimen collection and grouping Gastric mucosa tissues and archived wax lumps were collected from January 2016 to December 2018 in the department of pathology,affiliated hospital of Dali university.The medical history of all the included specimens was reviewed to ensure that the patients did not take nsaids,antibiotics or proton pump inhibitors within 4 weeks.The patients had no cardiopulmonary insufficiency,chronic liver disease,cirrhosis,chronic kidney disease or other diseases,and no history of other tumors.Paraffin sections were prepared from the specimens and stained with HE,and the pathological changes of gastric mucosal tissues were determined according to pathological diagnosis?superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia and gastric cancer?.At the same time,Helicobacte.pylori?Hp?infection was determined according to the 14C breath test by referring to the medical history.The specimens were divided into superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia,and gastric cancer groups with positive Hp infection and negative Hp infection.2 Detection of ASH2L and NF-?B signaling pathway activation related proteins The expressions of ASH2L,P65 and I?B?in superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia and gastric cancer were detected by immunohistochemistry?Elivision Super?.3 Statistical analysis The results of immunohistochemistry were compared by Fishers Exact Test,and the correlation was analyzed by Pearson correlation analysis.Results1 Fifteen cases of superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia and gastric cancer with positive and negative Hp infection were obtained.2 Test results of ASH2L?1?In the Hp positive group,ASH2L was gradually increased in superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia,and gastric cancer tissues,with positive rates of 6.7%,40.0%,53.3%,93.3%,and 100.0%,respectively.The protein positive expression rate in superficial gastritis difference was statistically significant compared with atrophic gastritis tissues,in atrophic gastritis and intestinal metaplasia tissues there was no statistically significant difference comparison,in intestinal metaplasia and hyperplasia tissues comparative difference was statistically significant,the hyperplasia there was no statistically significant difference compared with gastric cancer tissues.?2?In the Hp negative group,ASH2L was gradually increased in superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia,and gastric cancer tissues,and the positive rate was 13.3%,53.3%,80.0%,100.0%,and 100.0%,respectively.The protein positive expression rate in superficial gastritis difference was statistically significant compared with atrophic gastritis tissues,in atrophic gastritis and intestinal metaplasia tissues there was no statistically significant difference comparison,in intestinal metaplasia there was no statistically significant difference compared with hyperplasia tissues,and was consistent in dysplasia and gastric cancer tissues.?3?In the pathological changes of superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia and gastric cancer at various stages,there was no statistical significance in the positive expression rate of ASH2L between the Hp positive group and the negative group.?4?In addition to gastric mucosa epithelial cells,ASH2L can be expressed in lymphocytes,neutrophils and macrophages in the tissues.3 Test results of P65?1?In the Hp positive group,the expression of P65 in superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia,and gastric cancer gradually increased,and the positive rate was 13.3%,40.0%,60.0%,100.0%,and 100.0%respectively.The protein positive expression rate in superficial gastritis there was no statistically significant difference compared with atrophic gastritis tissues,in atrophic gastritis and intestinal metaplasia tissues there was no statistically significant difference comparison,in intestinal metaplasia and hyperplasia tissues comparative difference was the positive expression rate of P65 between the Hp positive group and the negative group.?4?Except gastric mucosa epithelial cells,lymphocytes,neutrophils and macrophages in the tissues all showed P65 expression.4 Test results of I?B??1?In the Hp positive group,the expressions of I?B?in superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia and gastric cancer were gradually increased,and the positive rates were 0,33.3%,40.0%,46.7%and 46.7%,respectively.The protein positive expression rate in superficial gastritis difference was statistically significant compared with atrophic gastritis tissues,in atrophic gastritis and intestinal metaplasia tissues there was no statistically significant difference comparison,in intestinal metaplasia there was no statistically significant difference compared with hyperplasia tissues,and was consistent in dysplasia and gastric cancer tissues.?2?In the negative group of Hp infection,the expressions of I?B?in superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia and gastric cancer were gradually increased,and the positive rates were 0,26.7%,40.0%,53.3%and 60.0%,respectively.The protein positive expression rate in superficial gastritis there was no statistically significant difference compared with atrophic gastritis tissues,in atrophic gastritis and intestinal metaplasia tissues there was no statistically significant difference comparison,in intestinal metaplasia there was no statistically significant difference compared with hyperplasia tissues,the hyperplasia there was no statistically significant difference compared with gastric cancer tissues.?3?In the pathological changes of superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia,and gastric cancer,there was no statistical significance in the positive expression rate of I?B?between the Hp positive group and the negative group.?4?The expression of I?B?was observed in lymphocytes,neutrophils and macrophages in the gastric mucosa in addition to epithelial cells.5 Correlation between ASH2L and P65 and I?B??1?In the positive Hp infection group and the negative Hp infection group,ASH2L positive expression rate and P65 positive expression rate were positively correlated in the pathological changes from superficial gastritis to gastric cancer.?2?In the positive Hp infection group and the negative Hp infection group,ASH2L positive expression rate and I?B?positive expression rate were positively correlated in the pathological changes from superficial gastritis to gastric cancer.Conclusion?1?NF-?B signaling pathway was activated in superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia,and gastric cancer in the Hp positive and Hp negative groups.?2?In the pathological changes of superficial gastritis,atrophic gastritis,intestinal metaplasia,dysplasia,and gastric cancer at various stages,there were no statistically significant differences in the expressions of ASH2L,P65,and I?B?between the positive and negative Hp infection groups.?3?It was found that the positive expression rate of ASH2L was positively correlated with the positive expression rate of P65,and the positive expression rate of ASH2L was positively correlated with the positive expression rate of I?B?,indicating that ASH2L was positively correlated with the activation of NF-?B signaling pathway.?4?The occurrence of gastric cancer is the result of the interaction of multiple factors.The NF-?B signaling pathway is activated under the action of multiple factors.Hp infection is not an independent risk factor for gastric cancer.Therefore,it is difficult to determine the relationship between ASH2L and NF-?B signaling pathway activation in Hp infection at histological level.
