Atherosclerosis(AS)and its complications are the leading cause of death in humans and they are one of the main risk factors for cardiovascular disease.A large number of studies have shown that atherosclerosis is a complex multi-factor chronic inflammatory Vascular disease,where Vascular Smooth Muscle Cell is the key factor(Abnormal proliferation of VSMC).Therefore,targeted therapy for abnormal proliferation of smooth muscle cells has become a hot spot in the treatment of atherosclerosis.Micrornas;Mirnas are small non-coding ribonucleic acids about 22 nucleotides in length.MiRNA is an efficient and specific gene regulator that regulates target proteins and participates in basic biological processes(including cell differentiation,proliferation,apoptosis and cell signaling pathways),and plays an important role in post-transcriptional regulation of genes.Caveolin protein-1(Caveolin-1;Cav-1)can inhibit tumor growth,regulate alveolar respiration,and play an important role in endocytosis and signal mediation.However,the effect of Cav-1 on VSMC in SD rats has not been reported.In order to investigate the role of Cav-1 protein in rat VSMC,primary smooth muscle cells of SD rats were cultured and identified in this study.SiRNA interference with the expression of Cav-1 was used to determine that Cav-1 had a negative regulatory effect on cell proliferation.Through TargetScan we matched Cav-1 with miR-204-5p.According to the research reports,microrna-204-5p(mir-204-5p)is an important factor regulating cell proliferation,which plays a role in regulating tumor cell proliferation in human breast tumor,mouse breast tumor and lymphoma,and also plays a role related to apoptosis in endothelial cells.However,whether mir-204-5p plays a role in VSMC and how it regulates VSMC function through specific target genes remains to be investigated.In order to further explore the correlation between mir-204-5p and Cav-1 and its role in VSMCs,mir-204-5p mimicks and inhibitors were transfected to detect the expression levels of related genes and proteins,cell activity,cycle,apoptosis and the expression of key pathway proteins.The AS model of SD rats was established and the gene and protein levels were measured.The above experiments proved that mir-204-5p targeted Cav-1 in VSMC of rats,and reduced the expression of Cav-1 protein,affected the proliferation activity of VSMC,and promoted the expression of PPI3K/PI3 K in the proliferation signaling pathway.The mechanism of mir-204-5p and its target gene Cav-1 in regulating VSMC function was preliminarily analyzed,providing a theoretical basis for mir-204-5p to regulate the related functions of rat VSMC by targeting Cav-1.
