Background and ObjectiveAs one of the most common intracranial malignant tumors,the incidence of glioma has been increasing in recent years,but its own infiltration makes it difficult to remove them.Except for a few low-grade gliomas with good prognosis after early active treatment,high-grade gliomas tend to have poor prognosis and low survival rate,which cause huge physiological and psychological burden to families and society.It is known that the mutation of IDH-1 and MGMT play an important role in the occurrence,development and radiotherapy and chemotherapy of glioma.The purpose of this paper is to further explore the expression of two markers in glioma patients with different pathological factors and the possible relationship between them,which provide some direction and guidance for the study of the pathogenesis and treatment of glioma.MethodNinety-two specimens of gliomas surgically resected from January 2015 to July2017 in Zhengzhou University were collected as experimental group.Brain tissue from 30 patients with craniocerebral trauma who underwent craniotomy and decompression at the same time was taken as control group.The inclusion criteria of the experimental group were as follows:preoperative MRI examination showed glioma,and pathological diagnosis was confirmed after operation;all the patients included were first onset,without radiotherapy,chemotherapy and surgical treatment.The inclusion criteria of the control group were:brain tissue obtained by craniotomy and decompression for trauma,and the brain tissues of non-functional areas confirmed during operation.Immunohistochemical method was used to detect the expression of IDH-1 and MGMT in brain tissues of the two groups.The expression of IDH-1 and MGMT in Western blot was analyzed by independent sample t-test.Combined with clinical and pathological factors and postoperative pathology,the expression of IDH-1 and MGMT in glioma tissues of the experimental group and the control group were analyzed by Max-2 test.Meanwhile,the expression of IDH-1 and MGMT in glioma tissues was further analyzed by Max-2 test in different pathological factors(age)of glioma patients.Gender,pathological grade,pathological type,location of glioma,epilepsy or not)and Speraman correlation analysis were used to analyze the relationship between the two markers.Result1.Expression of IDH-1 protein:The positive expression rate of IDH-1 was55.11%(54/98)in the experimental group and 16.7%(5/30)in the control group.There were differences between the two groups(?~2=13.656,P=0.0002).The expression of IDH-1 was not correlated with age,sex,type of case and location of glioma(P>0.05).The positive expression rate was 36.7%(22/60)in low-grade gliomas and 57.9%(22/38)in high-grade gliomas.There was a difference between them(?~2=4.238,P=0.0395).The positive expression rate was lower in gliomas with epilepsy(27/60)and higher in non-epilepsy tissues(25/38).Difference(?~2=4.45,P=0.0349).There were significant differences in IDH-1 expression between the two groups(t=-7.662,P=0.008),and between high-grade tumors and low-grade tumors(t=-6.673,P=0.009).2.Expression of MGMT protein:The positive expression rate of MGMT in the experimental group was 43.88%(43/98),while that in the control group was 13.3%(4/30).The expression of MGMT was not correlated with age,sex,pathological type,location of glioma and epilepsy(P>0.05).The positive rate was 45%(27/60)in low grade gliomas and 65.8%(25/38)in high grade gliomas.There was a difference between them(?~2=4.037,P=0.0445).There were significant differences in IDH-1expression between the two groups(t=-8.432,P=0.004),and between high-grade tumors and low-grade tumors(t=-7.761,P=0.007).3.The expression of IDH-1 and MGMT:There was a positive correlation between the expression of IDH-1 and MGMT(r=0.302,P=0.0049).Conclusion1.IDH-1 and MGMT protein are highly expressed in glioma tissues,and are higher in high-level than in low-level.,indicating that IDH-1 and MGMT play a role in the evolution of glioma.2.The expression of IDH-1 protein is closely related to the expression of MGMT protein.It is consistent with the expression of MGMT protein.It may have a potential connection in the occurrence and development of glioma and promote the progress of tumors.
