Effects And Possible Mechanism Of LncRNA-HOTAIR On Proliferation And Apoptotic Of Gastric Cancer Cells

Objective To investigate the effect of LncRNA-HOTAIR on proliferation and apoptosis of gastric cancer 7901 cells.Methods1.The human gastric cancer cells(SGC-7901)was divided into blank control group(Blank group),LncRNA-HOTAIR overexpression group and LncRNA-HOTAIR inhibition group(si-HOTAIR group).The blank group cells were not transfected with any vectors.The cells of LncRNA-HOTAIR overexpression group and si-HOTAIR group were transfected with HOTAIR overexpression plasmid and si-HOTAIR plasmid respectively.2.CCK8 and colony formation assay were used to detect the proliferation of cells.3.The apoptosis of cells was detected by flow cytometry and Caspase 3/7activity assay.4.qRT-PCR was used to detect the relative expression of LncRNA-HOTAIR and microRNA-126(miR-126)in each group.Results1.Compared with the Blank group,the LncRNA-HOTAIR overexpressiongroup cells showed a significant increase in proliferation and a decrease in apoptotic rate;The proliferation in the si-HOTAIR group cells was significantly inhibited while the apoptosis was promoted(p<0.05).2.There was a negative correlation between LncRNA-HOTAIR and miR-126 expression in gastric cancer cells(p<0.05);Conclusion LncRNA-HOTAIR could increase the proliferation and inhibite the apoptotic of gastric cancer 7901 cells,the mechanism may be related to the expression of mir-126 in gastric cancer cells.
