Objective:The hair follicle neural crest stem cells are derived from the skin’s hair follicles.which are located in the bulge of hair follicles and are a type of neural stem cell stem cells.Since most of the individual hair follicles are widely distributed,the source of the cells is relatively abundant.Also,the hair follicle neural crest is relatively easy to separate,and a large number of sufficient cell cultures can be performed.More importantly,hair follicle neural crest stem cells have many synaptic advantages compared to other adult stem cells,such as the advantages of good activity and low tumorigenicity in vivo.Hair follicle neural crest stem cells can differentiate into different types of cells under the stimulation of different inducing factors.For example,studies have been conducted to induce the cells into neuronal cells,glial cells,and smooth muscle cells.These induced cells not only changed their phenotype,but also changed their functions.Transplanting these induced cells into animals can also play a corresponding role.These characteristics of hair follicle neural crest stem cells become an ideal source of cells in cell therapy,tissue engineering,and drug development.Therefore,it is of great significance to conduct in-depth biological characteristics research on hair follicle stem cells.The largest number of glial cells in the central nervous system,and astrocytes,which account for the vast majority of them,play an important role in the central nervous system.Astrocyte dysfunction can cause many neurological diseases.However,people’s understanding of astrocyte function is mostly obtained through in vitro experiments.Therefore,in vitro culture of astrocytes plays a crucial role in the research.The astrocytes obtained by primary methods are more likely to be contaminated by other cells,which may have an effect on the research work of astrocytes.Therefore,the purpose of this experiment is to extract hfNCSCs from rats.After the extracted cells were cultured and identified,they were induced to differentiate into astrocytes by using a specific induction medium,and the inducing astrocytes were subjected to functional identification in vitro.Methods:(1)The cultured hair follicle neural crest stem cells were cultured using a tissue patch culture method,and then the cultured cells were subjected to immunofluorescence identification.(2)The extracted and cultured hair follicle neural crest stem cells were induced to induce differentiation into astrocytes,and the differentiated cells were identified by immunofluorescence staining.(3)In vitro functional detection of induced astrocytes:Induced astrocytes were used to detect the function of promoting neuron proliferation and synapse formation.Result:(1)The cells extracted and cultured from the hair follicle neural crest of rats have good activity,regular morphology and good value,and nestin immunofluorescence staining is positive.(2)The cells were induced to differentiate after culture,and the differentiated cells were positive for GFAP immunofluorescence staining,confirming that they were astrocytes.(3)Induced astrocytes can promote neuron cell proliferation and promote synapse formation.Conclusion:(1)Hair follicle neural crest stem cells can be cultured by tissue patch extraction.(2)The hair follicle neural crest stem cells can be induced to differentiate into astrocytes under specific induction conditions.(3)The induced astrocytes can survive,increase in value and have corresponding functions.
