BackgroundDiabetes mellitus(DM)is currently a common human metabolic disease characterized by hyperglycemia,which is caused by decreased insulin production by pancreatic cells and/or insulin resistance.In recent years,the incidence of diabetes has continued to rise and it has seriously affected human health and life.Nowadays,miRNA-26a(miR-26a-1,miR-26a-2)has gradually become a research hotspot.It can participate in the regulation of many conductive pathways and physiological processes of cells,and plays a very important role in the pathogenesis,development,and prognosis of many diseases.In recent years,experiments have found that liver miRNA-26a plays an important role in regulating carbohydrate metabolism,lipid metabolism,and insulin sensitivity by regulating key metabolic genes.Therefore,we speculate that in patients with type 2 diabetes mellitus(T2DM)and type 2 diabetes mellitus complicated with microvascular complications(T2DMC),the expression levels of miRNA-26a in peripheral blood may change and play an important role in the development of T2DM.PurposeTo investigate the relationship between plasma miRNA-26a levels and relatives metabolic markers in healthy people,patients with type 2 diabetes,and microvascular complications of type 2 diabetes mellitus and their clinical significance.Provides new ideas for the diagnosis of T2DM and provides new targets for the treatment of T2DM.Methods1.Subjects:Three groups were divided into simple type 2 diabetes(T2DM),type 2 diabetes microvascular complication(T2DMC)and healthy control group.A total of 32 patients with type 2 diabetes were included.Among them,9 patients with primary diabetes mellitus and 23 patients with diabetes mellitus were treated;30 patients with type 2 diabetes microvascular complications included 18 patients with diabetic nephropathy and 20 patients with diabetic retinopathy.There were 8 cases of diabetes and diabetic retinopathy;the healthy control group had 35 cases with normal physical examination indicators.2.Related Metabolic Indexes and General Data Collection:Fasting Blood Glucose(FPG),Fasting C-Peptide,HbAlc,Blood Pressure(BP),Triglyceride(TG),Total Cholesterol(TC),Low-Density Lipoprotein Cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),creatinine(SCr).Record the fundus situation.Carefully register body mass index,gender,age,current medical history,past history,family history,smoking,drinking,etc.3.The expression of plasma miRNA-26a was measured in all subjects using real-time quantitative PCR(qRT-PCR)4.Compare the expression levels of plasma miRNA-26a between the three groups,and analyze the correlation between the plasma miRNA-26a expression levels and related metabolic indicators,and investigate the clinical significance of plasma miRNA-26a expression levels in type 2 diabetes.5.Statistical analysis of the dataResult1.The expression of plasma miRNA-26a in patients with type 2 diabetes mellitus and type 2 diabetes microvascular complications was higher than that in healthy controls,and the difference was statistically significant(P<0.05)2.The expression of miRNA-26a in patients with type 2 diabetes microvascular complications was higher than that of patients with type 2 diabetes,but the difference was not statistically significant(P>0.05)3.Correlation analysis showed that plasma miRNA-26a expression levels were positively correlated with HbAlC,FPG,TC,TG,LDL-C and other metabolic indicators,negatively correlated with C peptides,and had no significant correlation with HDL-C,SCr(P>0.05)ConclusionCompared with the healthy control group,the expression of plasma miRNA-26a was significantly upregulated in patients with type 2 diabetes mellitus and type 2 diabetes microvascular complications.The expression level of plasma miRNA-26a is expected to become a potential new target for the diagnosis,treatment and prognosis of diabetes.
