Objective:To preliminarily explore the activity of benzothiazole derivative a10 against ovarian cancer SKOV3 cells,providing experimental basis for the research and development of new drugs for the treatment of ovarian cancer.Methods:?The effect of 14 synthetic benzothiazole derivatives on the MTT cell activity of human SKOV3 ovarian cancer cell lines was tested for 24 hours(cisplatin as a positive control).The IC50 values of 14 benzothiazole derivatives acting SKVO3 cell lines for 24 hours were calculated,and the compound a10 with the lowest IC50 value was used for subsequent experiments.?The effect of a10 on the apoptosis rate of SKOV3 cell line was detected by flow cytometry.We set up the experimental group(compound a10 concentration 25/50/75 mol/L),positive control group(cisplatin 0.66mol/L),and negative control group(same amount of high-sugar medium/0.9%normal saline).?The effect of compound a10 on the migration ability of SKOV3tumor cell lines was detected by cell scratch test and transwell test.?The long-term toxicity of a10 against the growth of human ovarian cancer SKOV3 tumor cell line was detected by 3D cell culture.?The mRNA expressions of bcl-2 and Bax of compound a10 against human ovarian cancer cell line SKOV3 at 8 h and 12 h were detected by real-time qPCR.?A model of chick embryo allantoic membrane(CAM)tumor was established and the inhibition of compound a10 on the growth of human ovarian cancer SKOV3 tumor cell line was tested.Results:Among the 14 benzothiazole derivatives,compound a10 had a prominent effect against human ovarian cancer SKOV3 tumor cell line,and its IC50 value of 72h was57.38?mol/L;The results of flow cytometry showed that high concentration of compound a10(experimental group)significantly promoted tumor cell apoptosis;The effect of compound a10 on the migration ability of SKOV3 tumor cell lines of human ovarian cancer was time-dependent and concentration-dependent;Compound a10 was detected by q-PCR to increase the mRNA expression levels of bcl-2 and Bax;The long-term inhibition of compound a10 on tumor cells was observed by 3D cell culture technology;Human ovarian cancer SKOV3 tumor cell line concentrate was planted on the chick embryo allantoic membrane after 8 days of incubation for3-5 days,and the SKOV3 cell number with the best tumor modeling rate was4*10~6,Under the microscope,tumor sections stained with HE had the same physiological characteristics as ovarian cancer tissues,The effect of a10 on the growth of SKOV3 cell tumor of human ovarian cancer was observed by the chick embryo allantoic membrane tumor model.Conclusion:Benzothiazole derivative a10 had better activity against human ovarian cancer SKOV3 cell line,which would become a novel compound against ovarian cancer.This provided a new research direction for the treatment of relapsed and drug-resistant ovarian cancer patients.
