Purpose:The mucosal-associated invarient T cells in peripheral blood of NPC patients were used to analyze the subtypes of MAIT cells in peripheral blood of NPC and its correlation with clinical progression of NPC patients.The role and significance of mucosal-associated invarient T cells in the diagnosis and treatment of NPC in peripheral blood were preliminarily described.Method:Byflowcytometry,theproportionandsubtypeclassificationof mucosa-associated invarient T cells in peripheral blood of healthy volunteers and NPC patients were compared and analyzed.the correlation between age and MAIT cell ratio was analyzed in the healthy volunteer group;To analyze the correlation between the proportion of mucosa-associated constant T cells in peripheral blood of patients with nasopharyngeal carcinoma and clinical indicators;Comparison of the secreted function of MAIT cells by vitro stimulated in two groups,preliminary analysis of the correlation between mucosal-associated constant T cells and NPC patients and its significance for the diagnosis and treatment of NPC;Result:1.There was a significant difference in the proportion of MAIT cells in CD3~+T cells between different age groups in healthy people(p<0.05),and it was found that MAIT cells accounted for CD3~+T in healthy adults by spearman correlation analysis.The proportion of cells was negatively correlated with age(r=-1.3679,p<0.05).The proportion of circulating MAIT cells in peripheral blood of patients with NPC was 0.57%,which was significantly lower than that of healthy controls(1.03%)(p<0.05).In a further subgroup analysis,it was found that most of the MAIT cells in peripheral blood of NPC patients were mainly CD8~+and DN(CD4~-CD8~-)subtypes,and CD4~+subtypes only accounted for a small part.CD8~+MAIT cells accounted for about 75.6%(45.20%-93.60%),DN subtypes accounted for 17.35%(1.10%-50.70%),and CD4~+subtypes only accounted for 2.62%(0.09%-13.30%),compared with healthy controls.There was no significant difference in comparison(p>0.05).2.The proportion of MAIT cells secreting IFN-?in peripheral blood of NPC patients was significantly higher than that of healthy controls(56.15%vs 31.80%,p<0.05),and the proportion of MAIT cells secreting TNF-?was also significantly higher than that of healthy controls(47.65%vs 13.85%,p<0.05).There was no significant difference in the ratio of MAIT cells secreting IL-2 and IL-17 in peripheral blood between NPC patients and healthy controls(4.34%vs 7.09%,p>0.05;1.85%vs 1.46%,p>0.05).3.The proportion of MAIT cells was significantly lower in peripheral blood of NPC patients(T3-4)with higher tumor T stage than in NPC patients(T1-2)with lower tumor T stage.There was no significant correlation between the ratio of MAIT cells to CD3~+T cells and lymph node metastasis or distant metastasis.There was no significant difference in the ratio of MAIT cells in peripheral blood of patients with serum EBV-DNA negative and serum EBV-DNA positive NPC.4.Compared with healthy controls,the proportion of MAIT cells expressing PD-1 in peripheral blood of NPC patients was significantly increased(10.06%vs15.10%,p<0.05).Conclusion:1.The proportion of MAIT cells in CD3+T cells in NPC patients was significantly lower than that in healthy controls.2.MAIT cells play an anti-tumor role in patients with nasopharyngeal carcinoma,and their functions are gradually depleted.3.MAIT cells may be a biological indicator for assessing the progression of NPC.4.The proportion of PD-1~+MAIT cells in peripheral blood of NPC patients was significantly up-regulated.
