Objective:To explore the role of macrophage migration inhibitory factor(MIF)on differentiation of pericyte in rabbit model of systemic sclerosis-associated pulmonary hypertension Methods:15 male rabbits were randomly divided into 3 groups: A(control group);B(model group);C(MIF inhibitor group).All groups were tested for right ventricular hypertrophy index(RVHI)and pulmonary artery pressure after modeling.The pathological changes and MIF expression in lung tissue were observed with HE staining and immunohistochemistry examination.Vascular pericytes were extracted from rabbit lung in three groups and the expressions of ?-SMA,and Col1A1 of pericytes were detected and compared by Western blotting.Results:(1)The right ventricular hypertrophy index and mean pulmonary artery pressure in group B and group C were significantly higher than those in group A(P<0.05).HE staining of dermis thickening,inflammatory cell infiltration in lung tissue and interstitial,pulmonary trunk and pulmonary small vessel wall Thickening,stenosis;(2)Immunohistochemistry showed that MIF in both groups B and C was highly expressed in lung tissue and located in the intima of the pulmonary arterioles.(3)The expression of ?-SMA in peripheral blood of group B and group C was significantly higher than that of group A(P<0.05).The expression of ?-SMA and Col1A1 in pericytes of group C was down-regulated compared with group B,and both were statistically significant(P< 0.05).Conclusion:(1)MIF is highly expressed in lung tissue of rabbit SSc-PAH model.(2)Decling of MIF activity inhibits differentiation of pericytes phenotype into myofibroblasts,and down-regulate the expression of ?-SMA and Col1A1 in pericytes.(3)MIF may participate in the pathological process of systemic sclerosis pulmonary hypertension by promoting the differentiation of perivascular cells into myofibroblasts.
