Diet is composed of a variety of foods and nutrients,different dietary patterns may affect individual physiological and pathological state.Therefore,we explored the effects of different dietary patterns on intestinal lactase activity and adipose tissue inflammation,which is important to further explore the effects of dietary patterns on body health and to explore the role of nutrients in food in the occurrence or intervention of diseases.Lactase activity was different in populations.Western populations have higher lactase activity,while Asian and African populations generally have lower lactase activity.Although many studies have shown that lactase activity was related to genetic factors,few papers have systematically studied the specific impact of dietary patterns on lactase activity.In this paper,we systematically studied the effects of low fat diet(LFD)and high fat diet(HFD)on lactase activity of contemporary and offspring male and female mice.On this basis,we used the others dietary patterns include the LFD and HFD added with lactose and the dietary restriction of LFD and HFD,to explore the effects of these diet on lactase activity.The results showed that:(1)the lactase activity of male mice were decreased after feeding of HFD,while the lactase activity of female mice was opposite to that of male mice.Feeding HFD in parental mice reduced lactase activity in offspring male mice,while lactase activity in offspring female mice did not change significantly as in male mice.(2)After lactose add with LFD,the lactase activity of male mice was significantly lower than that of the normal LFD group.After lactose add with HFD,the lactase activity of female mice was significantly lower than that of the normal HFD group.(3)The 20% and 40% dietary restriction of LFD was significantly reduced lactase activity in the small intestine of male mice.The occurrence of diet-related chronic diseases such as obesity can be prevented through the intervention of diet and lifestyles.It has been reported that dietary restriction can improve the occurrence of obesity and diabetes by affecting adipose tissue inflammation.Therefore,this paper also studied the effects of dietary restriction on adipose tissue inflammation in mice.It was found that dietary restriction significantly reduced the body weight and tissue weight of mice.The results of flow cytometry showed that dietary restriction could reduce the number of macrophages in the adipose tissue and the ratio of M1/M2 in mice fed with HFD.Quantitative PCR was performed to detect the mRNA levels of F4/80 and TNF? in mice EAT,and it was found that the inflammatory level in EAT decreased after dietary restriction.Dietary restriction in mice fed HFD can significantly reduce the content of TG,HDL and LDL in serum.It can enhance the ability of energy metabolism in mice fed HFD among the six hours before night.To sum up,we explored the effects of different dietary patterns,namely,dietary lipid content,lactose supplementation and different food intake on lactase activity in small intestine of mice.Dietary restriction could improve adipose tissue inflammation in obese mice,but could not completely restore to normal diet level.These results suggest that we should follow a reasonable diet pattern in life,so as to maintain a healthy state of the body.
