Objective To observe the protective effect of resveratrol on renal injury induced by iohexol in diabetic rats,and to explore its mechanism.Methods There were 45 SPF type 2 diabetic rats with a body mass of 250-300 g.Renal injury model of diabetic rats was established by intra-arterial injection of iohexol.The randomized digital table method was used to randomly divide into three groups: sham operation group(Sham group,n=15) and iohexol group(NL group,n=15).And the resveratrol group(Res group,n=15).The Res group was given resveratrol(100 mg/kg) before the modelling,and the NL group and the Sham group were given the same amount of normal saline for 5 days.On the 6th day,the rats were fasted for water,the 7th day.Rats were intraperitoneally injected with 2% sodium pentobarbital anesthesia,fixed in supine position,stripped femoral vein,puncture femoral vein,NL group and Res group were injected with indomethacin(10 mg/kg),L-nitrosamine Methyl ester(10 mg/kg),iohexol 3 gI/kg(slow injection within 10 min),and Sham group were injected with the same amount of normal saline at the same time.After 48 hours of model establishment,rats were sacrificed.Serum creatinine(Scr) and blood urea nitrogen(BUN) concentrations were observed in each group.Renal pathological changes were observed by HE staining.Total superoxide dismutase(T-SOD) in serum was detected by ELISA.Malondialdehyde(MDA) and glutathione peroxidase(GPX) concentrations;TUNEL detection of renal epithelial cell apoptosis;q RT-PCR,Western blot detection of SIRT1,HIF-1? protein expression.Results Compared with the Sham group,the renal tubular epithelial cells in the NL group were vacuolar degeneration,the cells fell off into the small cavity,the tubular structure was destroyed,the renal tubular epithelial cells were apoptotic,and the total plasma Scr,BUN and MDA were significantly increased.T-SOD GPX was significantly decreased(P<0.05).The expression of SIRT1 was significantly decreased in the kidney group,while the expression of HIF-1? was significantly increased(P<0.05).Resveratrol can alleviate renal damage in diabetic rats induced by iohexol.Compared with NL group,renal tubular epithelial cells in Res group are less damaged,only a small amount of renal tubular cells are apoptotic,and total plasma T-SOD and GPX are obvious.When elevated,Scr,BUN and MDA were significantly decreased(P<0.05).The expression of HIF-1? was significantly decreased in the kidney group,while the expression of SIRT1 was significantly increased(P <0.05).Conclusions Resveratrol had a protective effect on rat model of kidney injury induced by iohexol,which may be related to the regulation of SIRT1/HIF-1? signaling pathway.
