Purpose : To study the expression of KRT1 and PI3 K in breast carcinoma tissues and to investigate the relationship of their expression with clinical pathological characteristics and prognosis of breast cancer.Methods:1.The database of Kyoto Encyclopedia of Genes and Genomes(KEGG)is employed to analyze the bioinformatics for KRT1.First,KAAS,the online software service tool of LEGG database,is used to annotate the submitted KRT1.Then the annotated KRT1 is matched to the corresponding pathway in database by KEGG mapper analysis tool for analysis.2.The immunohistochemical method is used to test the expression of KRT1 protein and PI3 K protein in 50 cases of mammary cancer and paracancerous tissue.The statistical method is used to analyze the difference of both in expression of mammary cancer and paracancerous tissue,and the relation between both and clinicopathological features of mammary cancer & prognosis.Pearson is used to analyze the expression correlation between and KRT1 protein and PI3 K protein in mammary cancer tissue.3.The molecular hybridization in situ is used to test the expression of KRT1 RNA in mammary cancer and paracancerous tissue.And Pearson is used to analyze the expression correlation between KRT1 RNA and KRT1 protein in mammary cancer tissue.Results:1.Using KEGG database KRT1 bioinformatics analysis found that breast cancer associated with ER signaling pathways,Estradiol in ER signaling pathways combined with ER after on the one hand can regulate gene transcription,participate in KRT1 expression of regulation,on the other hand to adjust the c-Src protein involved in PI3K/AKT signaling pathway regulating;2.(1)The positive rates of KRT1 protein in mammary cancer and paracancerous tissue are 62.0%(31#50)and 32.0%(16#50)respectively,with statistical significance in difference(P<0.05).The expression of KRT1 protein is related to the ER and PR status of patients with mammary cancer,as well as whether there is recurrence after 2 years of operation(both P<0.05),in which positive expression rate(70.3%)of KRT1 in ER positive mammary cancer is significantly higher than that of ER negative mammary cancer(38.5%).The positive expression rate(74.3%)of KRT1 in PR positive mammary cancer is higher than that of PR negative mammary cancer(33.3%).The positive expression rate(43.8%)of KRT1 in mammary cancer recurred after 2 years of operation is significantly lower than mammary cancer recurred after 2 years of operation(88.0%).KRT1 protein in mammary cancer is not significantly related to age of patients with mammary cancer,tumor size,lymphatic metastasis or not,tissue classification,different HER-2 and Ki67 proliferation index(All P>0.05);(2)The positive rates of PI3 K protein in mammary cancer and paracancerous tissue are76.0%(38#50)and 24.0%(12#50)respectively,with statistical significance in difference(P<0.05).The expression of PI3 K protein is related to the ER status of patients with mammary cancer,as well as whether there is recurrence after 2 years of operation(both P<0.05),in which positive expression rate(67.6%)of PI3 K in ER positive mammary cancer is significantly lower than that of ER negative mammary cancer(100.0%).The positive expression rate(87.5%)of PI3 K in mammary cancer recurred after 2 years of operation is significantly higher than mammary cancer recurred after 2 years of operation(52.0%).PI3 K protein in mammary cancer is not significantly related to age of patients with mammary cancer,tumor size,lymphatic metastasis or not,tissue classification,PR status,different HER-2 and Ki67 proliferation index(AllP>0.05);(3)the expression of KRT1 protein is negatively correlated with the expression of PI3 K protein in mammary cancer tissue(r=-0.343,P < 0.05).3.The result of molecular hybridization in situ shows that the positive rates of KRT1 m RNA in mammary cancer and paracancerous tissue are 68.0%(34#50)and 24.0%(12#50)respectively,with statistical significance in difference(P<0.005).In mammary cancer,KRT1 m RNA is located in endochylema and nucleus.The signal intensity is not significantly changed from cancerous center to edge.The expression of KRT1 m RNA is positively correlated with that of KRT1 pertain in mammary cancer(r=0.435,P < 0.05).Conclusion: 1.The expression of KRT1 in mammary cancer is significantly up-regulated and is related to ER status and the expression of PI3 K.ER may,together with PI3K/AKT signaling pathway,produce effect on prognosis of patients with mammary cancer through participation in the expression of KRT1.2.KRT1 may turn into a potential biomarker related to prognosis of mammary cancer,providing clues for the further exploration of nosogenesis about mammary cancer.
