Objective: To prepare ITP animal model by injecting rat anti-mouse CD41 monoclonal antibody(MWReg30)into Balb/c mice,and to study the intervention with QiShun BaoLiEr.Peripheral blood platelet,observe the ITP model mice bone marrow megakaryocyte,spleen index,platelet associated antibody and platelet surface PAIg G monoclonal antibody CD41/CD61 and so on,To explore the mechanism of QiShun BaoLiEr in the treatment of ITP at animal level.Methods: Rat anti-mouse CD41 monoclonal antibody(MWReg 30 monoclonal antibody)was injected into the abdominal cavity of Balb/c mice for 7 days.At 0,24,72,120 and 168 hours after injection,the tail vein blood of mice was taken to detect the number of platelets.When the platelet level dropped to 0,24,72,120 and 168 hours after injection,the platelet level was measured.When the normal(pre-modeling)level is less than half,it is confirmed that the modeling is successful.After the success of ITP model in Bal B/c mice,90 mice were randomly divided into six groups according to platelet count: normal control group,model group,QiShun BaoLiEr low-dose group,QiShun BaoLiEr medium-dose group,QiShun BaoLiEr high-dose group and positive control group.There were 15 rats in each group.Normal control group and model group were given saline intragastrically at 0.2ml/10 g body weight for 7 days before modeling,once a day for 7 days.The low dose group(0.4g/kg),the middle dose group(1.2g/kg)and the high dose group(2.0g/kg)were given different concentrations of drug suspension respectively.The positive control group was given Xuekang capsule(1.2g/kg)concentration of drug suspension once a day,and the mice were intraperitoneally injected with CD41 monoclonal antibody for 7 days.After the experiment,the general state of mice and platelet count,the number of megakaryocytes in bone marrow,spleen and thymus index,platelet-related antibody PAIg G,platelet surface specific antibody CD41/CD61 were observed to study the pharmacological mechanism of QiShun BaoLiEr in the treatment of ITP.Results: 1.Establishment of ITP mouse model:The results showed that thrombocytopenia could be induced by intraperitoneal injection of rat anti-mouse CD41 monoclonal antibody(MWReg30).The number of platelets in the model group decreased to about 40% of the original platelets 72 hours after injection,indicating that the establishment of ITP mouse model was basically successful.The results were as follows: 1.Observation of appearance and behavior of mice in each group: mice in the normal group had good mental state,action and reaction,glossy fur and normal diet;mice in the model group had poor mental state,less activity with piles,dark fur and less diet;mice in the QiShun BaoLiEr group and the control group had good mental state,no obvious changes in diet,glossy fur and normal activity.2.There was no significant difference in body weight among the groups(P > 0.05).3.Comparisons of platelet count in mice of each group: Compared with normal control group,platelet count in peripheral blood of model group mice decreased significantly,with significant difference(P < 0.05).Compared with the model group,the platelets in each group of QiShun BaoLiEr increased significantly(P < 0.05).4.Compared with the model group,the number of megakaryocytes in bone marrow of mice in QiShun BaoLiEr group increased significantly(P < 0.05).5.Observation of organ index of mice in each group: Compared with normal mice,spleen index of model mice was significantly higher than that of normal mice(P < 0.05).Compared with the model group,the spleen index of mice in QiShun BaoLiEr group decreased significantly,with significant difference(P < 0.05).There was no significant difference in thymus index among groups(P > 0.05).6.Platelet related antibody PAIg G of mice in each group: compared with the normal group,the platelet related antibody PAIg G of the model group was significantly increased,with a statistically significant difference(P<0.05).Compared with the model group,the platelet-associated antibody PAIg G was significantly reduced in the QiShun BaoLiEr group(P<0.05).7.Expression of CD41/CD61,a platelet surface specific antibody,in each group: compared with the model group,the expression of CD41/CD61,a platelet surface specific antibody,was significantly increased in the QiShun BaoLiEr group(P<0.05).Conclusion:1.The method of preparing ITP animal model by injecting rat anti-mouse CD41 monoclonal antibody(MWReg30)is basically successful,but this method has no obvious effect on the general state,signs and thymus of mice in a short time.2.Mongolian medicine QiShun BaoLiEr may improve immune system function by reducing platelet-related antibodies,increasing peripheral platelets and reducing organ index in mice.3.Mongolian medicine QiShun BaoLiEr may promote platelet production by promoting the differentiation and maturation of platelet-producing megakaryocytes,regulating the expression of platelet surface-specific antibody CD41/CD61,thus achieving the effect of ITP treatment.
