Objective: To investigate the association between single nucleotide polymorphism(SNP)of rs4796793 in STAT3 gene and genetic susceptibility as well as prognosis of bladder cancer.Methods: In the present study,TaqMan probe real-time PCR technique was used to analyze SNP rs4796793 in STAT3 gene in bladder cancer patients and controls.Chi-square(?2)test was used to compare the genotype and allele frequencies between two groups.Odds ratio(OR)and 95% confidence intervals(CI)were calculated using logistic regression.The log-rank test and Cox regression analysis were used to evaluate the association between genotypes of SNP with patients' outcome from the date of primary diagnosis until recurrence or death.Results: SNP rs4796793 was successfully genotyped in 306 patients with bladder cancer and 525 control subjects.Our results indicated that the genotype and allele frequencies of SNP rs4796793 were significantly different between the bladder cancer patients and normal subjects(OR=1.36,95%CI: 1.11-1.68,p=0.003).In addition,stratified analyses and survival analysis revealed that the polymorphism of rs4796793 was also associated with bladder patients' recurrence rate and overall-survival(NMIBC patients)(p<0.05).Conclusion: These findings suggested that polymorphism of rs4796793 in STAT3 gene was associated with the genetic susceptibility,recurrence rate and overall-survival of bladder cancer in the Chinese Han population.
