Objective: To investigate the role and mechanism of USP20 in breast cancer,we observed the changes of biological behavior of breast cancer cell MCF-7 and its possible relationship with ANXA2,UBE3 A and P53 by shRNA-USP20 lentivirus knockdown.Methods: MCF-7 breast cancer cells were transfected with shRNA-USP20 lentivirus,and the shRNA-USP20 interference fragment with the highest interference efficiency was screened by q-PCR and Western blot for subsequent experiments.CCK8-kit was used to detect the proliferation of cells after knocking down USP20.flow cytometry was used to detect the apoptosis rate after knocking down USP20.Transwell chamber test was used to detect the migration and invasion ability of cells after knocking down USP20;and finally,q-PC was used to detect the migration and invasion ability of cells after knocking down USP20.The expression of ANXA2,UBE3 A and p53 genes in breast cancer cells knocked down by USP20 was detected by q-PCR and Western blot.Results: In knockdown group(KD),the expression of USP20 was significantly decreased(P = 0.0067),and the expression of USP20 protein was also significantly decreased(P = 0.001).CCK8 assay showed that the proliferation of MCF-7 cells was significantly inhibited after knockdown of USP20(P< 0.05).Flow cytometry showed that the apoptosis rate was significantly increased after knockdown of USP20(P = 0.028).Transwell chamber experiment showed that the migration and invasion of MCF-7 cells after knockdown of USP20 were significantly inhibited(P< 0.05).After knocking down USP20,the expression of p53 gene and p53 protein decreased significantly(P< 0.0001),but there was no significant difference in the expression of ANXA2 and UBE3A(P> 0.05).Conclusion: USP20 may play an important role in the occurrence and development of breast cancer.Its mechanism may be closely related to p53,but not to UBE3 A and ANXA2.
